鹽脅迫對南方泡桐基因表達(dá)的影響

【摘要】:為闡明泡桐多倍體的耐鹽分子機(jī)制,以南方泡桐二倍體及對應(yīng)的四倍體為試驗材料,以白花泡桐基因組為參考序列,利用RNA-Seq測序技術(shù),研究了鹽處理前后南方泡桐四倍體和對應(yīng)二倍體的基因表達(dá)變化。通過比對分析,共鑒定出與鹽脅迫相關(guān)的差異表達(dá)基因2 940個。對這些差異基因進(jìn)行GO功能分類,結(jié)果顯示差異基因共參與了39個分類,集中在細(xì)胞、結(jié)合、催化等分類功能區(qū)的數(shù)量最多。KEGG代謝通路分析發(fā)現(xiàn),這些差異表達(dá)基因主要參與了Plant hormone signal transduction,Carbon metabolism,Biosynthesis of amino acid等,其中編碼MYB、WRKY、bZIP、ATPase等的基因差異表達(dá)顯著。表明這些基因可能是潛在的鹽脅迫響應(yīng)基因。采用qRTPCR技術(shù)驗證了隨機(jī)挑選的9個差異表達(dá)基因,結(jié)果與轉(zhuǎn)錄組測序數(shù)據(jù)趨勢一致。
[Abstract]:In order to elucidate the molecular mechanism of salt tolerance of Paulownia paulownia polyploid, the diploid and tetraploid of Paulownia paulownia were used as experimental materials, the genome of Paulownia paulownia was used as reference sequence, and the RNA-Seq sequencing technique was used. The changes of gene expression of paulownia tetraploid and diploid were studied before and after salt treatment. A total of 2940 differentially expressed genes related to salt stress were identified by comparative analysis. The GO functional classification of these differentially expressed genes showed that the differentially expressed genes were involved in 39 classifications, mainly in cell, binding, catalytic and other functional regions. KEGG metabolic pathway analysis showed that, These differentially expressed genes were mainly involved in Plant hormone signal transduction,Carbon metabolism,Biosynthesis of amino acid et al. Among them, the differentially expressed genes encoded MYB,WRKY,bZIP,ATPase et al. These genes may be potential salt stress response genes.
