【摘要】：通過超景深顯微鏡和掃描電鏡觀察不同發(fā)育階段的佛手(Citrus medica L.var.sarcodactylis Swingle)花芽形態(tài),發(fā)現佛手指狀部分并非發(fā)育自柱頭,而是來自雌蕊子房。同時,在佛手中分離了CmsSUN20、CmsOFP7、CmsOFP12和CmsYABBY5等4個果形相關基因的全長,與甜橙序列比對的相似性大于95%,構建進化樹分析發(fā)現其與甜橙聚在一個分支。收集佛手和香櫞(C.medica L.)果實發(fā)育相同階段不同組織(葉片,花瓣,雌蕊,雄蕊)材料,以qRT-PCR法分析了上述4個基因的表達。結果表明CmsSUN20在佛手雌蕊的表達顯著高于香櫞,在其他組織中差異不顯著或相反,表明該基因可能與佛手雌蕊發(fā)育有關。CmsYABBY5在佛手雌蕊和雄蕊中都顯著高表達,而在葉片中卻顯著低表達,表明CmsYABBY5有可能對佛手雌蕊和雄蕊發(fā)育有重要作用。而CmsOFP7和CmsOFP12在佛手葉片、雌蕊和雄蕊中均高表達,由于并非特異在雌蕊中高表達,所以可能并不對佛手果形建成起決定作用。
[Abstract]:The floral bud morphology of (Citrus medica L.var.sarcodactylis Swingle in different stages of development was observed by means of hyper-depth microscope and scanning electron microscope. It was found that the finger part of the Buddha developed not from the stigma but from the pistil ovary. At the same time, four fruit shape related genes, CmsSUN20,CmsOFP7,CmsOFP12 and CmsYABBY5, were isolated from the phalanx, and the similarity of alignment with orange sequence was greater than 95. The phylogenetic tree analysis showed that it was clustered in a branch with sweet orange. C.medica L. The expression of the four genes in different tissues (leaves, petals, pistil, stamens) at the same stage of fruit development was analyzed by qRT-PCR method. The results showed that the expression of CmsSUN20 in pistil was significantly higher than that in Citron, but there was no significant difference in other tissues, suggesting that the expression of CmsSUN20 might be related to the development of pistil. CmsYABBY5 was significantly overexpressed in both pistil and stamen of Vertex. However, the expression of CmsYABBY5 was significantly lower in leaves, suggesting that CmsYABBY5 may play an important role in the development of pistil and stamen. However, CmsOFP7 and CmsOFP12 were highly expressed in the leaves, pistil and stamen, but they were not specifically expressed in the pistil, so they may not play a decisive role in the fruit formation.
