轉(zhuǎn)錄組與蛋白質(zhì)組關聯(lián)分析鑒定桑樹硬枝扦插皮部生根相關功能基因

【摘要】:前期以桑品種育711號硬枝扦插皮部生根的插穗為材料,對扦插生根過程起始階段(S0)、根原基形成階段(S1)和不定根伸長階段(S2)的插穗基部皮層進行了轉(zhuǎn)錄組與蛋白質(zhì)組測序。在此基礎上對差異表達基因與蛋白質(zhì)(差異系數(shù)1.5,錯誤發(fā)現(xiàn)率≤0.05)做關聯(lián)分析。結(jié)果顯示:不同階段插穗基部皮層的基因和蛋白質(zhì)表達趨勢總體上呈正相關,關聯(lián)系數(shù)分別為0.470 3(S0-vs-S1)與0.113 0(S1-vs-S2);基因與蛋白質(zhì)表達趨勢相反的關聯(lián)數(shù)量較少,關聯(lián)系數(shù)分別為-0.319 9(S0-vs-S1)與-0.107 1(S1-vs-S2);基因與蛋白質(zhì)表達趨勢相同的關聯(lián)數(shù)量相對較多,關聯(lián)系數(shù)分別為0.855 4(S0-vs-S1)與0.834 6(S1-vs-S2),均呈強相關。在關聯(lián)分析表達趨勢相同組中篩選鑒定出21個差異表達基因,并進行功能注釋與信號通路分析。其中,有5個基因在生根過程中持續(xù)上調(diào)表達,主要包括酚氧化酶(PPO)、馬鈴薯糖蛋白(Patatin)、生長素酰胺水解酶(ILL)、肌氨酸氧化酶(SOX)的編碼基因;有13個基因在根原基形成期上調(diào)表達,在不定根伸長期下調(diào)表達,主要包括赤霉素雙加氧酶(GA3OX4)、12-氧植物二烯酸還原酶(OPR)、內(nèi)切葡聚糖酶(EG)和吲哚乙酸氨基化合成酶(GH3)的編碼基因;有3個基因在根原基形成期下調(diào)表達,在不定根伸長期上調(diào)表達,分別為細胞膜相關鈣結(jié)合蛋白(PCa P)、肌肉LIM蛋白(MLP)和膜聯(lián)蛋白(ANX)的編碼基因。上述關聯(lián)分析的結(jié)果為深入認識桑樹插穗皮部生根過程的生理機制提供了相關功能基因信息。
