蕎麥脫落酸不敏感基因序列分析

【摘要】:脫落酸不敏感基因(ABA-insensitive,ABI)是調(diào)控植物逆境脅迫應(yīng)答、種子脫落和休眠等的一類(lèi)轉(zhuǎn)錄因子。生物信息學(xué)分析發(fā)現(xiàn),普通蕎麥ABI(CL38501)基因c DNA全長(zhǎng)為963 bp,編碼320個(gè)氨基酸,含有典型的b ZIP結(jié)構(gòu)域和ABI5家族保守區(qū)Ⅱ,推測(cè)該基因可能編碼ABI5家族蛋白。與野生種質(zhì)相比,花蕾期和開(kāi)花期栽培品種ABA信號(hào)轉(zhuǎn)導(dǎo)途徑中絕大多數(shù)基因表達(dá)量上調(diào),其中,ABI基因表達(dá)量分別上調(diào)4.42和3.66倍。對(duì)9個(gè)栽培品種和5份野生種質(zhì)的ABI基因進(jìn)行PCR擴(kuò)增,發(fā)現(xiàn)14份材料的405個(gè)位點(diǎn)中多態(tài)位點(diǎn)為30個(gè),9個(gè)栽培品種間僅含1個(gè)多態(tài)位點(diǎn),5個(gè)野生種質(zhì)間含有7個(gè)多態(tài)位點(diǎn),說(shuō)明普通蕎麥ABI基因序列高度保守。栽培品種與野生種質(zhì)ABI序列存在11個(gè)共同氨基酸差異位點(diǎn),蛋白構(gòu)象預(yù)測(cè)發(fā)現(xiàn)第32位點(diǎn)和第60位點(diǎn)氨基酸發(fā)生置換導(dǎo)致蛋白構(gòu)象發(fā)生變化,表明其可能與栽培馴化過(guò)程中蕎麥喪失休眠與落粒特性相關(guān)。
