轉(zhuǎn)基因大豆MON89788基體標(biāo)準(zhǔn)物質(zhì)的研制

【摘要】:轉(zhuǎn)基因產(chǎn)品成分檢測是轉(zhuǎn)基因生物安全管理和標(biāo)識的重要支撐,而轉(zhuǎn)基因生物標(biāo)準(zhǔn)物質(zhì)是獲得準(zhǔn)確、可靠、具有可比性檢測結(jié)果的保證。轉(zhuǎn)基因大豆(Glycine max)MON89788為我國批準(zhǔn)進(jìn)口用作加工原料的農(nóng)業(yè)轉(zhuǎn)基因生物,因此,制備轉(zhuǎn)基因大豆MON89788基體標(biāo)準(zhǔn)物質(zhì)是對其進(jìn)行安全管理所必需的。本研究通過將轉(zhuǎn)基因大豆MON89788和其受體材料A3244分別冷凍研磨成粉末,按質(zhì)量分?jǐn)?shù)稱量配制,得到轉(zhuǎn)基因含量為49.3 g/kg的標(biāo)準(zhǔn)物質(zhì)。采用方差分析法(F檢驗(yàn)法)進(jìn)行均勻性檢驗(yàn),統(tǒng)計(jì)量F值為1.618,小于臨界值F_((0.05,11,24)),標(biāo)準(zhǔn)物質(zhì)均勻性良好;短期穩(wěn)定性考察結(jié)果表明,標(biāo)準(zhǔn)物質(zhì)可在常溫下穩(wěn)定儲存4周,在-20℃下長期穩(wěn)定性達(dá)到30個(gè)月。本批MON89788基體標(biāo)準(zhǔn)物質(zhì)有兩個(gè)特性量值,一是基于重量法配比混合的粉末質(zhì)量分?jǐn)?shù),量值為49.3 g/kg,擴(kuò)展不確定度為2.0 g/kg;二是基于q RT-PCR 8家實(shí)驗(yàn)室聯(lián)合定值的轉(zhuǎn)基因DNA與總DNA的質(zhì)量分?jǐn)?shù),量值為49.8 ng/μg,擴(kuò)展不確定度為5.9 ng/μg。本批標(biāo)準(zhǔn)物質(zhì)每瓶1.0 g,使用時(shí)最小取樣量為100 mg,可用于對轉(zhuǎn)基因大豆MON89788進(jìn)行定性和定量檢測,以及轉(zhuǎn)基因大豆MON89788特異性檢測方法的評價(jià)和實(shí)驗(yàn)室質(zhì)量控制。本研究為我國轉(zhuǎn)基因檢測基體標(biāo)準(zhǔn)物質(zhì)的研制提供了理論基礎(chǔ)和技術(shù)支持。
[Abstract]:Component detection of transgenic products is an important support for the safety management and marking of transgenic organisms, and the standard material of transgenic organisms is the guarantee of obtaining accurate, reliable and comparable test results. Genetically modified soybean (Glycine max) MON89788) is an agricultural transgenic organism approved by China to be used as raw material for processing. Therefore, the preparation of MON89788 matrix reference material is necessary for its safety management. In this study, the transgenic soybean MON89788 and its receptor material A3244 were ground into powder by freezing, and the standard material with transgenic content of 49.3 g/kg was obtained according to the weight fraction. Using the method of variance analysis (F test) to test the homogeneity, the statistic F value is 1.618, which is less than the critical value F _ (0.05 ~ 1124),). The results of short-term stability test showed that the standard materials could be stored steadily for 4 weeks at room temperature and 30 months at -20 鈩,
