肥胖型和瘦肉型豬的脂肪、肝臟及骨骼肌組織中CIDE家族基因表達(dá)水平的比較研究(英文)

【摘要】:目的:研究CIDE家族基因在肥胖型和瘦肉型豬的脂肪、肝臟及肌肉組織中的基因表達(dá)水平差異,并初步探CIDE家族基因與脂質(zhì)代謝的關(guān)系。創(chuàng)新點:首次在肥胖型與瘦肉型豬模型中解釋CIDE家族基因可以調(diào)節(jié)脂質(zhì)代謝,并有助于脂肪沉積及導(dǎo)致肥胖。方法:采用熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(qP CR)檢測肥胖型藍(lán)塘豬和瘦肉型杜長大豬的脂肪、肝臟和骨骼肌中CIDE家族基因、SREBP-1c、PGC-1α、HNF-4α、FASN、DGAT1和DGAT2、perlipin 2等基因表達(dá)水平。采用血漿生化指標(biāo)儀試劑盒檢測兩個品種豬血漿中甘油三酯、葡萄糖、游離脂肪酸及膽固醇的含量。結(jié)論:肥胖型藍(lán)塘豬脂肪和背最長肌組織中的Cidea和Cidec,及肝臟中Cidec的基因表達(dá)量明顯高于瘦肉型杜長大豬。在脂肪組織中,脂質(zhì)代謝相關(guān)的基因(包括SREBP-1c、PGC-1α、HNF-4α、FASN、DGAT1和DGAT2基因)表達(dá)量都是藍(lán)塘豬高于杜長大豬。藍(lán)塘豬肝臟中的SREBP-1c、HNF-4α和PGC-1α基因表達(dá)水平顯著高于杜長大豬。藍(lán)塘豬背最長肌組織的SREBP-1c、HNF-4α、PGC-1α和DGAT2基因表達(dá)量高于杜長大豬。然而,藍(lán)塘豬的脂肪、肝臟及背最長肌三種組織中的perlipin 2的表達(dá)量顯著低于杜長大豬。此外,藍(lán)塘豬血漿中的甘油三酯、葡萄糖及游離脂肪酸濃度明顯高于杜長大豬。通過相關(guān)性分析,我們發(fā)現(xiàn)肥胖型和瘦肉型豬不同組織中的CIDE家族基因表達(dá)水平與背部脂肪厚度、腹部脂肪重量、血漿中的甘油三酯、葡萄糖及游離脂肪酸濃度有明顯的正向相關(guān)性。綜上所述,CIDE家族基因可以調(diào)節(jié)脂質(zhì)代謝,并促進(jìn)脂肪沉積及導(dǎo)致肥胖。
[Abstract]:Aim: to study the difference of gene expression of CIDE family gene in fat, liver and muscle tissues of obese pigs and lean pigs, and to explore the relationship between CIDE family genes and lipid metabolism. Innovation: for the first time, CIDE family genes can regulate lipid metabolism, contribute to fat deposition and lead to obesity in obese and lean pig models. Methods: fluorescence quantitative polymerase chain reaction (qP CR) was used to detect CIDE family genes, SREBP-1c,PGC-1 偽, HNF-4 偽, FASN,DGAT1 and DGAT2, in fat, liver and skeletal muscle of obese and lean Landrace pigs. Perlipin 2 gene expression level. The contents of triglycerides, glucose, free fatty acids and cholesterol in plasma of two breeds of pigs were determined by plasma biochemical indicator kit. Conclusion: the expression of Cidea and Cidec, in fat and longissimus dorsi muscle tissues and Cidec gene expression in liver of obese blue tang pigs were significantly higher than those of lean Duroc pigs. In adipose tissue, the expression levels of genes related to lipid metabolism (including SREBP-1c,PGC-1 偽, HNF-4 偽, FASN,DGAT1 and DGAT2 genes) were higher in Lantang pigs than in du grown-up pigs. The expression levels of SREBP-1c,HNF-4 偽 and PGC-1 偽 in the liver of Lantang pig were significantly higher than those in Du-grown pig. The expression of SREBP-1c,HNF-4 偽, PGC-1 偽 and DGAT2 genes in the longissimus dorsi muscle tissue of Lantang pig was higher than that in Du-grown pig. However, the expression of perlipin 2 in the fat, liver and longissimus dorsi of Lantang pigs was significantly lower than that of Duroc pigs. In addition, the concentrations of triglycerides, glucose and free fatty acids in the plasma of Lantang pigs were significantly higher than those in Duroc pigs. By correlation analysis, we found that the expression level of CIDE family genes in different tissues of obese and lean pigs was related to the thickness of back fat, weight of abdominal fat, triglyceride in plasma. The concentrations of glucose and free fatty acids were positively correlated. In conclusion, CIDE family genes regulate lipid metabolism, promote fat deposition and lead to obesity.
