安徽農(nóng)村地區(qū)HIV感染者和幼兒園兒童藍(lán)氏賈第鞭毛蟲感染情況及其基因型
[Abstract]:Objective to investigate and compare the (Giardia lamblia) infection and genotype of Giardia lamblia in rural areas of Anhui province. Methods from April 24 to May 9, 2015, we collected the relevant information of HIV infected persons registered at an AIDS treatment site in rural Anhui and all children in a local kindergarten, and collected feces for iodine staining and microscopic examination. Fecal DNA, was extracted from positive individuals. Nested PCR was used to amplify and sequence the triosephosphate isomerase,tpi gene of Giardia lamblia. Clustal X 1.83 and MEGA 6.0 were used to compare the homology and phylogenetic analysis. Results A total of 127 cases of HIV infection and 125 cases of children were investigated. Four cases of Giardia lamblia infection were found, including 3 cases of children and 1 case of HIV infection. The infection rate was 2.40% (3 / 125) and 0.79% (1 / 127), respectively. There was no significant difference between the two groups (P0.05). The feces of 3 children infected with infection were soft stool, no diarrhea, abdominal pain and other clinical symptoms. The feces of one case of HIV infection were dilute stool with obvious abdominal pain, diarrhea, fatigue and weight loss. The results of PCR detection showed that the fecal DNA amplification products of 4 cases of HIV infected patients had specific bands at about 500 bp. The sequencing results showed that all the 4 cases were infected with Giardia lamblia. The sequence similarity of two repeat samples of HIV infection was 79, and the similarity was 79 with the reported (Gen Bank accession number: KF271445 of Shanghai Giardia lamblia isolates. 98. The similarity of the three infected children was 99g, and the similarity with Shanghai Giardia flagellate isolate was 79g. The results of phylogenetic tree constructed by tpi gene showed that the isolates of Giardia lambtii infected with HIV were compound agglomeration A BU. All the isolates of 3 children infected with HIV were agglomerates. There was a high homology between the agglomeration A of HIV infection and the isolated strains of 3 children infected with HIV. Conclusion there is Giardia lamblia infection in HIV infected persons and children in rural areas of Anhui province. The genotype of HIV infected individuals is compound agglomeration A B, and that of children is agglomeration A.
