胃癌患者Vav1基因D324Y位點多態(tài)性檢測的意義

【摘要】：目的分析胃癌患者外周血Vav1基因D324Y位點多態(tài)性及白細(xì)胞介素-6(IL-6)、腫瘤標(biāo)志物的關(guān)系,對其意義進(jìn)行探討。方法選取2013年1月-2015年12月確診胃癌的患者300例為病例組,同時取100例體檢健康者為對照組。化學(xué)發(fā)光法測定血清腫瘤標(biāo)志物CA199水平;酶聯(lián)免疫法(ELISA)檢測外周血Vav1及IL-6水平;直接測序技術(shù)檢測Vav1基因D324Y位點多態(tài)性情況。結(jié)果病例組外周血中Vav1、CA199、IL-6水平均高于對照組(P0.05)。病例組中Vav1基因D324Y位點TT、GT的基因型頻率分別為7.0%、19.3%,高于對照組(0%、5.0%)(P0.05);病例組中T等位基因的頻率為16.67%,高于對照組(2.5%)(P0.05)。兩因素方差分析結(jié)果顯示,IL-6、CA199、Vav1的水平與患病狀態(tài)及基因型均有關(guān)系(P0.001)。Logistic回歸結(jié)果顯示TT基因型與胃癌關(guān)系密切,是胃癌的獨立危險因素(P0.05)。結(jié)論 Vav1基因D324Y位點多態(tài)性可能是胃癌發(fā)生進(jìn)展的重要影響因素。
[Abstract]:Objective to analyze the relationship between Vav1 gene D324Y polymorphism, interleukin-6 (IL-6) and tumor markers in peripheral blood of patients with gastric cancer. Methods 300 patients with gastric cancer from January 2013 to December 2015 were selected as case group and 100 healthy persons as control group. The levels of serum tumor marker CA199, peripheral blood Vav1 and IL-6 were detected by chemiluminescence assay, and the polymorphism of D324Y locus of Vav1 gene was detected by direct sequencing. Results the level of Vav1,CA199,IL-6 in peripheral blood of the case group was higher than that of the control group (P0.05). The genotypic frequencies of TT,GT at D324Y locus of Vav1 gene in the case group were 7. 0 and 19. 3%, respectively, which were higher than those in the control group (0. 0%) (P0.05). The frequency of T allele in the case group was 16.67%, which was higher than that in the control group (2.5%) (P0.05). Two-factor ANOVA showed that the level of IL-6,CA199,Vav1 was correlated with the status and genotype of gastric cancer (P0.001). Logistic regression analysis showed that TT genotype was closely related to gastric cancer and was an independent risk factor for gastric cancer (P0.05). Conclusion the polymorphism of D 324Y locus of Vav1 gene may be an important factor in the development of gastric cancer.
