綿羊OAS1基因PCR的優(yōu)化條件

【摘要】:為了研究綿羊OAS1基因片段的PCR最佳反應(yīng)條件,試驗(yàn)采集綿羊外周血液從中分離出白細(xì)胞,并對OAS1基因片段的PCR擴(kuò)增條件進(jìn)行優(yōu)化,通過對引物量、退火溫度、模板c DNA的純度進(jìn)行篩選,建立最優(yōu)反應(yīng)體系。結(jié)果表明:PCR反應(yīng)的最佳cDNA量為2μL,最佳上、下游引物量為1μL,最佳退火溫度為60℃。采用上述條件獲得的PCR產(chǎn)物,OAS1基因片段的目的條帶清晰、明亮,非特異性產(chǎn)物較少。
[Abstract]:In order to study the optimal PCR reaction conditions of sheep OAS1 gene fragments, the white blood cells were isolated from sheep peripheral blood, and the PCR amplification conditions of OAS1 gene fragments were optimized. The purity of template c DNA was screened and the optimal reaction system was established.
