‘云香’水仙NAC基因克隆及表達(dá)分析

【摘要】:從‘云香’水仙轉(zhuǎn)錄組文庫中篩選出1條NAC基因序列,采用PCR技術(shù)克隆得到其c DNA序列。序列分析表明,該NAC基因包含822 bp的開放閱讀框,編碼273個(gè)氨基酸,分子量30 903.9 u,命名為Nt NAC3153(Gen Bank登錄號:KU375569)。理化性質(zhì)分析和多重序列比對顯示,Nt NAC3153蛋白是一個(gè)無跨膜區(qū)域的親水性蛋白,在N端具有一段保守結(jié)構(gòu)域。系統(tǒng)進(jìn)化樹分析表明,Nt NAC3153與同為單子葉植物的小果野芭蕉Ma NAC、二穗短柄草Bd NAC和小米Si NAC親緣關(guān)系最近。實(shí)時(shí)熒光定量PCR分析顯示,Nt NAC3153基因能夠被ABA、鹽脅迫和高溫誘導(dǎo)表達(dá)。研究表明:Nt NAC3153可能參與‘云香’水仙的抗逆調(diào)控機(jī)制。
[Abstract]:A NAC gene sequence was screened from the transcriptional library of Narcissus sinensis, and its c DNA sequence was cloned by PCR technique. Sequence analysis showed that the NAC gene contained an open reading frame of 822 bp, encoding 273 amino acids with a molecular weight of 30 903.9 us.It was named Nt NAC3153 (Gen Bank accession number: KU375569). Physicochemical properties and multiple sequence alignment showed that Nt NAC3153 protein was a transmembrane free hydrophilic protein with a conserved domain at the N-terminal. Phylogenetic tree analysis showed that Nt NAC3153 had the closest relationship with Bd NAC and Si NAC of Ma NAC, and Si NAC, both monocotyledonous plants. Real time fluorescence quantitative PCR analysis showed that Nt NAC3153 gene could be expressed under ABA, salt stress and high temperature. It is suggested that Nt NAC3153 may be involved in the stress resistance regulation mechanism of Narcissus sinensis.
