貴州省結(jié)核分枝桿菌臨床分離株MIRU-VNTR位點(diǎn)基因分型研究

【摘要】:目的了解貴州省結(jié)核分枝桿菌臨床分離株的數(shù)目可變串聯(lián)重復(fù)序列(MIRU-VNTR)基因多態(tài)性及近期傳播情況。方法收集2014年貴州省結(jié)核分枝桿菌臨床分離株185株(耐多藥菌株57株,全敏感菌株97株),采用MIRU-VNTR 12個(gè)位點(diǎn)分析法進(jìn)行PCR檢測(cè),應(yīng)用NTsys2.10軟件對(duì)全部基因型數(shù)字化結(jié)果進(jìn)行非加權(quán)組平均法(UPGMA)聚類分析。結(jié)果 185株結(jié)核分枝桿菌臨床分離株分為5個(gè)基因群(Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴ),呈現(xiàn)156個(gè)基因型,其中135個(gè)為獨(dú)特基因型,其余50株為16簇,最大簇包含10株菌,最小簇包含2株菌,成簇率為27.03%,近期感染最小估計(jì)為18.37%,I群結(jié)核分枝桿菌為147株,占79.46%,為優(yōu)勢(shì)菌群,在MIRU 12個(gè)位點(diǎn)中,26、31位點(diǎn)多態(tài)性較高(h0.60),4、10、23、27、39、40位點(diǎn)呈中等程度的多態(tài)性(0.3≤h≤0.60),2、16、20、24位點(diǎn)多態(tài)性較低(h0.30)。結(jié)論貴州省結(jié)核分枝桿菌具有基因多態(tài)性,其主要的流行群為I群,MIRU26和MIRU31呈高等程度多態(tài)性,MIRU2、MIRU20和MIRU24、MIRU27呈低等程度多態(tài)性。
