黃牡丹PlWDR3和PlWDR18轉(zhuǎn)錄因子基因的克隆與表達(dá)
[Abstract]:Using Yunnan wild yellow peony as experimental material, 19 Unigene sequences with high homology of WDR40 protein related to anthocyanin synthesis were obtained according to the Unigene screening of WDR40 protein provided by the established transcriptional database of petals of Yunnan wild yellow peony, and named PlWDR1~19.. By amino acid sequence comparison and phylogenetic tree analysis, it was found that PlWDR3 and PlWDR18 may be involved in the regulation of anthocyanin synthesis in Peony. The ORF of PlWDR3 contains an open reading frame of 1 032bp and encodes a 344-amino acid protein. The ORF with the similarity of 83.29 and PlWDR18 contains an open reading frame of 1 035bp and encodes a protein of 1 345 amino acids, which is the most similar to the grape VvWDR2, reaching 93.77. Relative fluorescence quantitative PCR analysis showed that the expression patterns of PlWDR3 in different stages of yellow peony and purple peony were basically similar, the expression level of PlWDR3 was the highest in yellow peony and purple peony round peach, and the lowest in the initial opening stage, and PlWDR18 showed an upward trend in the early development of yellow peony. It reached the highest peak in the permeation period, then decreased, and reached the minimum value in the initial blooming stage, and increased in the early stage of the development of purple peony, reached the maximum value in the round peach stage, and then decreased gradually. The expression of PlWDR18 in yellow peony and purple peony was higher than that of PlWDR3. It was speculated that PlWDR3 and PlWDR18 were involved in the regulation of yellow peony color formation, which laid a foundation for further study on the mechanism of yellow peony color formation.
