茶樹(shù)HD-Zip轉(zhuǎn)錄因子基因的克隆及其對(duì)非生物脅迫的響應(yīng)分析
[Abstract]:As a kind of leaf plant, tea plant has important economic value. Abiotic stress has a great effect on the growth and development of tea plants and tea production. In this study, a gene encoding HD-Zip transcription factor, CsHB1., was cloned from tea plant 'Longjing 43'cDNA by RT-PCR method. Sequence analysis showed that the cDNA length of CsHB1 gene was 1 380 bp, encoding 459 amino acids, which contained a typical START domain. Phylogenetic analysis showed that tea CsHB1 belonged to the IV subfamily of transcription factors of HD-Zip family. The amino acid sequence of CsHB1 was 78.05% similar to that of other species, such as grape, tobacco, sesame and so on. The physicochemical properties, hydrophobicity and disordering of CsHB1 transcription factors showed that CsHB1 transcription factors were hydrophobic proteins and alkalescent, and there was no disorder region. The spatial structure analysis shows that CsHB1 has three 偽 -helix, many 尾 -folds and START domain. The expression of CsHB1 in high temperature (38 擄C), low temperature (4 擄C), drought (200g L ~ (-1) NaCl (200 mmol L ~ (-1) treatment for 812 h was analyzed by fluorescence quantitative PCR. The results showed that CsHB1 gene could be induced to express under different abiotic stress, and the difference of expression was significant.
