基于SMRT測序技術(shù)的16S rRNA基因全長測序及其分析方法
[Abstract]:Single molecular sequencing, known as the third generation sequencing technology, is a high throughput sequencing technology developed in recent years. The monomolecular real-time sequencing technology (SMRT) developed by Pacbio Bio Sciences Company is the first commercial technology. SMRT sequencing technology successfully overcomes the disadvantage of low accuracy of the third generation sequencing technology by cyclic sequencing of template sequence to produce ring uniform sequence (CCS),. With SMRT sequencing, scientists can explore the structure and function of microbes in complex environments more accurately. This paper introduces the advantages and disadvantages of SMRT sequencing technology in microbial 16s rRNA gene sequencing, and summarizes the important analysis links such as quality control, error sequence elimination, clustering and annotation analysis of full-length 16s rRNA gene sequence obtained from SMRT sequencing technology. At the same time, the possible problems and solutions of using SMRT sequencing technology to study microbes in complex environment are put forward, which are expected to provide reference for researchers.
