蝴蝶蘭Rubisco活化酶基因PhRCAα的序列特征及在低溫脅迫下的表達分析

【摘要】:為研究蝴蝶蘭Rubisco活化酶基因(RCA)及其在低溫脅迫下的響應(yīng)機制,本研究利用RT-PCR及RACE技術(shù)從蝴蝶蘭葉片中克隆了一個Rubisco活化酶基因PhRCAα的c DNA序列(登錄號KU187968)該基因全長1 642 bp,編碼473個氨基酸,包含P-loop_NTPase超家族結(jié)構(gòu)域,預(yù)測其成熟蛋白的分子質(zhì)量為46.16 k D,等電點為5.73,屬于RCA基因的α亞基。實時熒光定量PCR分析顯示,在13℃/8℃的晝夜溫度條件下,蝴蝶蘭PhRCAα基因的轉(zhuǎn)錄表達在處理3 d、6 d、9 d和15 d時明顯下降,恢復(fù)正常溫度條件后,該基因的表達升高;在4℃低溫條件下,PhRCAα基因的轉(zhuǎn)錄表達在處理0.5 h、1 h和2 h內(nèi)逐漸升高,而在處理4 h時其表達量下降,一直到低溫處理48 h時,其表達量維持在較低水平。結(jié)果表明PhRCAα對馴化低溫(13℃/8℃)和冷脅迫(4℃)具有不同的響應(yīng)機制,PhRCAα對短期的冷脅迫有一定的防御作用。
[Abstract]:To study the mechanism of Rubisco activase gene (RCA) and its response to low temperature stress in Phalaenopsis. In this study, the c DNA sequence of Rubisco activase gene PhRCA 偽 (accession number KU187968) was cloned from the leaves of Phalaenopsis by RT-PCR and RACE techniques. The whole length of 1 642 bp, encodes 473 amino acids, including P-loop_NTPase superfamily domain. The molecular weight of the mature protein was 46.16 KD and the isoelectric point was 5.73, which belonged to the 偽 subunit of RCA gene. Real-time fluorescence quantitative PCR analysis showed that the transcriptional expression of PhRCA 偽 gene in Phalaenopsis was significantly decreased at 3 days, 6 days, 9 days and 15 days after treatment at 13 鈩,
