線粒體COⅠ基因在觀賞魚中物種鑒定與分類應用

【摘要】:觀賞魚種類繁多,形態(tài)各異,市場上多是以商品名命名,在物種鑒定分類上存在一定的難度。本研究基于線粒體COⅠ基因,探討其在觀賞魚中物種鑒定與分類應用的可行性。通過PCR擴增,獲取18種觀賞魚249個個體的COⅠ基因序列;贙imura雙參數(shù)模型,利用MEGA軟件計算18種觀賞魚種間和種內(nèi)遺傳距離,并構建系統(tǒng)進化樹。結果顯示,觀賞魚類的種間遺傳距離明顯大于種內(nèi)遺傳距離,線粒體COⅠ基因序列可以用于物種鑒定。在系統(tǒng)進化樹中,系統(tǒng)進化關系跟傳統(tǒng)分類方法所得數(shù)據(jù)一致性高。研究表明,觀賞魚線粒體COⅠ基因序列,在物種鑒定與進化分類上具有一定的應用價值。
