小麥3B短臂染色體抗赤霉病主效QTL區(qū)域候選基因的表達(dá)

【摘要】:通過研究赤霉病菌接種后的基因表達(dá),篩選小麥赤霉病抗性相關(guān)基因。根據(jù)前人研究結(jié)果,利用FGENESH軟件對(duì)小麥3B短臂抗赤霉病主效QTL所在的小麥基因組測(cè)序獲得的重疊群ctg0954核苷酸序列進(jìn)行基因預(yù)測(cè),共預(yù)測(cè)得到489個(gè)基因。以攜帶該QTL的蘇麥3號(hào)和未攜帶該QTL的揚(yáng)麥15為材料,在小麥花期進(jìn)行赤霉病菌接種誘導(dǎo)基因表達(dá),接種后0 h、4 h、8 h、12 h、24 h對(duì)其中有功能注釋的259個(gè)預(yù)測(cè)基因的表達(dá)進(jìn)行半定量分析,獲得27個(gè)在品種間差異表達(dá)的基因。這27個(gè)基因的表達(dá)分為4類:一直上調(diào)表達(dá)(Ⅰ型),在接種后的早期上調(diào)表達(dá)后期下調(diào)表達(dá)(Ⅱ型),接種后早期下調(diào)表達(dá)后期上調(diào)表達(dá)(Ⅲ型),接種后一直下調(diào)表達(dá)(Ⅳ型)。在差異表達(dá)基因中,選擇6個(gè)基因,進(jìn)行Real-time PCR分析,結(jié)果表明這些基因在接種病原菌后均上調(diào)表達(dá),且在蘇麥3號(hào)和揚(yáng)麥15號(hào)品種間存在基因表達(dá)差異,驗(yàn)證了半定量表達(dá)結(jié)果,可作為與赤霉病抗性相關(guān)3B短臂主效QTL的候選基因。
