基于HRM體系開(kāi)發(fā)抗稻瘟病基因Pi2特異性分子標(biāo)記

【摘要】:Pi2是一個(gè)具有廣譜抗性的稻瘟病抗性基因,利用分子標(biāo)記輔助選擇技術(shù)選育含有Pi2抗性基因的水稻品種已得到越來(lái)越廣泛的應(yīng)用。本研究根據(jù)稻瘟病抗性基因Pi2及其復(fù)等位基因的序列,利用HRM技術(shù)開(kāi)發(fā)與Pi2完全共分離的分子標(biāo)記,并結(jié)合水稻DNA快速提取方法,建立中高通量輔助選擇體系。該方法體系準(zhǔn)確高效、簡(jiǎn)便快速、成本低廉,能夠大大提高抗病育種選育的效率,為分子標(biāo)記輔助選擇育種或者水稻種質(zhì)資源Pi2基因型鑒定提供技術(shù)選擇。
[Abstract]:Pi2 is a broad-spectrum resistant rice blast resistance gene. The selection of rice varieties with Pi2 resistance gene by molecular marker-assisted selection technology has been more and more widely used. Based on the sequence of rice blast resistance gene Pi2 and its complex alleles, a molecular marker completely separated from Pi2 was developed by using HRM technique, and a medium and high throughput assisted selection system was established by combining with the rapid extraction method of DNA in rice (Oryza sativa L.). The method is accurate, efficient, simple, rapid and low cost. It can greatly improve the efficiency of breeding for disease resistance, and provide a technical choice for molecular marker-assisted selection or Pi2 genotyping of rice germplasm resources.
