白血病慢粒急變過程相關(guān)基因的生物信息及臨床分析

【摘要】:急性粒細(xì)胞白血病(AML)是一種白細(xì)胞異常生長并積累從而干擾正常血細(xì)胞生長的惡性腫瘤。迄今為止,急性粒細(xì)胞白血病是如何由慢性粒細(xì)胞白血病轉(zhuǎn)化的機(jī)制仍不清楚。本研究中,我們發(fā)現(xiàn)長鏈非編碼RNA FAM30A在慢性粒細(xì)胞白血病(CML)的加速期和急變期均表達(dá)較高,而在CML的慢性期表達(dá)較低。在對(duì)急性粒細(xì)胞白血病的患者的臨床流行病學(xué)分析中,FAM30A的表達(dá)量是與總生存時(shí)間顯著相關(guān)的預(yù)后因子,且在多變量分析時(shí),當(dāng)調(diào)節(jié)了多個(gè)危險(xiǎn)因素后,FAM30A過表達(dá)仍與患者生存時(shí)間呈顯著負(fù)相關(guān)。進(jìn)一步研究表明,在年齡小于60歲的AML患者和正常核型的AML患者中,FAM30A高表達(dá)的病人預(yù)后均較差。我們的研究結(jié)果表明,FAM30A可作為急性粒細(xì)胞白血病的一個(gè)獨(dú)立預(yù)后因子,將為研究慢粒急粒變病理機(jī)制提供重要理論根據(jù)。
[Abstract]:Acute myeloid leukemia (AML) is a malignant tumor with abnormal growth and accumulation of leukocytes, which interferes with the growth of normal blood cells. So far, the mechanism of transformation of acute myeloid leukemia from chronic myeloid leukemia remains unclear. In this study, we found that the expression of long chain noncoding RNA FAM30A was higher in the accelerated and acute phase of chronic myeloid leukemia (CML), but lower in the chronic phase of CML. In the clinical epidemiological analysis of patients with acute myeloid leukemia, the expression of FAM30A was a prognostic factor significantly related to the total survival time, and in multivariate analysis, After adjusting multiple risk factors, the overexpression of FAM30A was negatively correlated with survival time. Further studies showed that the prognosis of patients with high expression of FAM30A was poor in both AML patients under 60 years old and AML patients with normal karyotype. Our results suggest that FAM30A may be an independent prognostic factor for acute myeloid leukemia, which will provide an important theoretical basis for the study of the pathological mechanism of chronic granulocytosis.
