磷酸酶Wip1基因敲除表型及機(jī)制研究進(jìn)展

【摘要】:野生型p53誘導(dǎo)的磷酸酶1(Wild-type p53-induced phosphatase1,Wip1)是PP2C(The type 2Cfamily of protein phosphatases)磷酸酶家族中的一員。在DNA損傷修復(fù)中起重要作用,并作為原癌基因受到越來越多的關(guān)注。Wip1敲除小鼠雖然能夠存活,但壽命縮短,并出現(xiàn)多系統(tǒng)異常,包括生殖系統(tǒng)、免疫體系、內(nèi)分泌系統(tǒng)、神經(jīng)系統(tǒng)等,具體表現(xiàn):雄性生殖器官萎縮及雄性不育、對病原體敏感、T細(xì)胞和B細(xì)胞功能受損、胰島素抵抗、體重增長受限、脂肪量減少、焦慮和抑郁樣行為增加等。相關(guān)機(jī)制研究發(fā)現(xiàn),Wip1通過去磷酸化不同蛋白底物而在多種生理和病理過程中發(fā)揮重要作用。本文旨在對小鼠Wip1基因缺失所致表型及其初步的分子機(jī)制進(jìn)行綜述,為進(jìn)一步研究Wip1的生物學(xué)功能以及哺乳動(dòng)物體內(nèi)生理和病理過程的發(fā)生機(jī)制提供參考。
[Abstract]:Wild-type p53-induced phosphatase 1 (Wip1) induced by wild type p53 is a member of PP2C (The type 2Cfamily of protein phosphatases) phosphatase family. Wip1 knockout mice, which play an important role in the repair of DNA damage, have received more and more attention as proto-oncogenes. Although Wip1 knockout mice can survive, their lifespan is shortened, and multiple system abnormalities occur, including reproductive system, immune system, endocrine system. Nervous system, specific manifestations: male reproductive organ atrophy and male sterility, sensitive to pathogens T cell and B cell function damage, insulin resistance, body weight growth limited, decreased fat, anxiety and depression like behavior increase. It has been found that Wip1 plays an important role in many physiological and pathological processes by dephosphorylation of different protein substrates. The aim of this paper is to review the phenotypes and molecular mechanisms of Wip1 gene deletion in mice, and to provide references for further study on the biological functions of Wip1 and the mechanism of physiological and pathological processes in mammals.
