【摘要】：目的:觀察樂胃飲對胃癌前病變(PLGC)大鼠胃黏膜中自噬體及其相關基因Beclin1的影響。方法:將60只雄性Wistar大鼠分為正常組,模型組,維酶素組,樂胃飲低、中、高劑量組,采用2%水楊酸鈉灌胃、饑飽失常以及自由飲用1-甲基-3-硝基-1-亞硝基胍(MNNG)溶液等綜合因素誘發(fā)PLGC模型。給藥28d后,利用透射電鏡觀察大鼠胃黏膜細胞自噬體,通過RT-PCR檢測自噬基因Beclin1 m RNA的表達。結果:與模型組比較,樂胃飲各劑量組胃黏膜Beclin1 m RNA表達顯著升高(P0.05,P0.01),電鏡可觀察到自噬體。結論:樂胃飲可能通過提高Beclin1的表達,增強PLGC胃黏膜自噬而降低癌變的可能性。
[Abstract]:Aim: to observe the effect of Lue Wei Yin on autophagy and its related gene Beclin1 in gastric mucosa of (PLGC) rats with gastric precancerous lesion. Methods: sixty male Wistar rats were divided into normal group, model group, retinol group, low, middle and high dosage group of lewei Yin, which were perfused with 2% sodium salicylate. The PLGC model was induced by combined factors such as hunger and satiety and free drinking of 1-methyl-3-nitro-1-nitrosoguanidine (MNNG) solution. After 28 days of administration, the autophagy of rat gastric mucosal cells was observed by transmission electron microscope, and the expression of Beclin1 m RNA was detected by RT-PCR. Results: compared with the model group, the expression of Beclin1 m RNA in gastric mucosa of each dose group was significantly higher than that in the model group (P0. 05 and P0. 01). Autophagy was observed under electron microscope. Conclusion: Lue Wei Yin may decrease the possibility of carcinogenesis by increasing the expression of Beclin1 and enhancing PLGC gastric mucosal autophagy.
