羅勒花和葉的轉(zhuǎn)錄組數(shù)據(jù)組裝及基因功能注釋

【摘要】:目的系統(tǒng)了解羅勒Ocimum basilicum花和葉的轉(zhuǎn)錄組特征,豐富羅勒轉(zhuǎn)錄組數(shù)據(jù)信息。方法選取羅勒新鮮花朵和葉片為材料,利用Illumina Hi SeqTM 2500進(jìn)行高通量測序,構(gòu)建羅勒花和葉轉(zhuǎn)錄組文庫,并用測序評(píng)估、轉(zhuǎn)錄組數(shù)據(jù)組裝和基因功能注釋等生物信息學(xué)方法進(jìn)行分析。結(jié)果對原始數(shù)據(jù)進(jìn)行除雜之后,共獲得86 331 137個(gè)reads片段,包含了6 455 365 309個(gè)核苷酸序列信息;將質(zhì)控后得到的高質(zhì)量序列進(jìn)行de novo拼接,得到了90 341個(gè)Unigene片段。將Unigene與COG數(shù)據(jù)庫進(jìn)行比對表明,羅勒的花和葉轉(zhuǎn)錄組中的Unigene根據(jù)功能可大致分為25類;根據(jù)GO功能分類可大致分為生物過程、細(xì)胞組分和分子功能3大類43分支;利用KEGG數(shù)據(jù)庫作為參考,依據(jù)代謝通路可以將轉(zhuǎn)錄組中的數(shù)據(jù)分成111類,包括生化代謝通路、植物病原體互作、DNA剪切、植物激素生物合成、苯丙氨酸生物合成、萜類化合物與類固醇類化合物合成、脂類代謝、RNA降解等。MISA軟件成功設(shè)計(jì)15 617對SSR引物,檢測到10 254個(gè)SNP位點(diǎn)。結(jié)論研究結(jié)果為后期羅勒功能基因的挖掘、代謝途徑及調(diào)控機(jī)制的研究奠定了理論基礎(chǔ)。
[Abstract]:Objective to study the transcriptional characteristics of basil Ocimum basilicum flowers and leaves, and to enrich the information of basil transcriptional data. Methods using Illumina Hi SeqTM 2500 as a high throughput sequencing method, a library of basil flower and leaf transcriptome was constructed and evaluated by sequencing. Bioinformatics methods such as transcriptional data assembly and gene function annotation were analyzed. Results after removing the original data, 86 331 137 reads fragments were obtained, including 6 455,365,309 nucleotide sequences, and 90 341 Unigene fragments were obtained by de novo splicing of high quality sequences obtained by quality control. The comparison of Unigene and COG database showed that Unigene in flower and leaf transcriptome of basil could be divided into 25 classes according to function, 43 branches according to go functional classification, 3 kinds of biological process, cell component and molecular function. Using KEGG database as reference, the data in transcriptome can be classified into 111 categories according to metabolic pathway, including biochemical metabolic pathway, plant pathogen interaction DNA cleavage, plant hormone biosynthesis, phenylalanine biosynthesis. A total of 15 617 pairs of SSR primers were successfully designed by MISA software, such as synthesis of terpenoids and steroids, lipid metabolism and RNA degradation, and 10 254 SNP sites were detected. Conclusion the results provide a theoretical basis for the excavation of basil functional genes and the study of metabolic pathways and regulatory mechanisms.
