【摘要】：目的觀察Hp對(duì)丙肝肝硬化患者外周血Foxp3、RORγt基因及相關(guān)細(xì)胞因子表達(dá)的影響,初步探討Hp感染對(duì)丙肝肝硬化患者Treg細(xì)胞調(diào)控作用的影響。方法選取丙肝肝硬化合并Hp感染患者42例為觀察組,未合并Hp感染患者30例為對(duì)照組,RT-PCR法測(cè)定基因表達(dá),ELISA法檢測(cè)相關(guān)細(xì)胞因子含量。結(jié)果觀察組Fox P3、RORγt基因相對(duì)表達(dá)水平顯著高于對(duì)照組;觀察組IL-10、IL-6、IL-17、TGF-β1、IFN-γ表達(dá)水平高于對(duì)照組,IL-2表達(dá)水平低于對(duì)照組;兩組患者Foxp3基因的表達(dá)與IL-10、TGF-β水平呈正相關(guān),與IL-2水平呈負(fù)相關(guān)。結(jié)論 Hp可能通過誘導(dǎo)Treg細(xì)胞的高表達(dá),促進(jìn)IL-10、TGF-β1、IL-6、IL-17的高表達(dá),抑制IL-2、IFN-γ表達(dá),造成體內(nèi)Treg/Th17、Th1/Th2平衡紊亂,促進(jìn)丙肝肝硬化患者肝細(xì)胞炎癥及纖維化進(jìn)程。
[Abstract]:Objective to observe the effect of HP on the expression of Foxp3 ror 緯 t gene and related cytokines in peripheral blood of patients with cirrhosis of hepatitis C, and to explore the effect of HP infection on the regulation of Treg cells in patients with cirrhosis of hepatitis C. Methods 42 patients with hepatitis C cirrhosis complicated with HP infection were selected as observation group and 30 patients without HP infection as control group. Results the relative expression level of ROR 緯 t gene in the observation group was significantly higher than that in the control group, and the expression level of IL-10, IL-6, TGF- 尾 1, IFN- 緯 in the observation group was higher than that in the control group, and the expression of Foxp3 gene was positively correlated with the level of IL-10 TGF- 尾 and negatively correlated with the level of IL-2. Conclusion HP may induce the overexpression of Treg cells, promote the high expression of IL-10 TGF- 尾 1 and IL-6N- 緯, inhibit the expression of IL-2 IFN- 緯, cause the imbalance of Treg-Th17 Th1 / Th2, and promote the progression of hepatocyte inflammation and fibrosis in patients with cirrhosis of hepatitis C.
