棉花抗逆基因GhAREB4的克隆及功能分析

【摘要】:AREBs轉(zhuǎn)錄因子家族基因主要參與干旱、高鹽、低溫等脅迫應(yīng)答反應(yīng),在植物抵御各種逆境脅迫中起著非常重要的作用。該研究經(jīng)序列電子拼接克隆了陸地棉GhAREB4基因,該基因全長(zhǎng)1 784bp,其開放閱讀框?yàn)? 227bp,編碼408個(gè)氨基酸,預(yù)測(cè)分子量為44.3kD,等電點(diǎn)為8.88。蛋白結(jié)構(gòu)預(yù)測(cè)發(fā)現(xiàn),該蛋白二級(jí)結(jié)構(gòu)中含有bZIP基因家族的保守結(jié)構(gòu)域。系統(tǒng)進(jìn)化樹分析表明,GhAREB4與可可的AREB轉(zhuǎn)錄因子同源性最高。綠色熒光蛋白亞細(xì)胞定位分析表明,GhAREB4蛋白分布在細(xì)胞核內(nèi)。qRT-PCR分析表明,GhAREB4基因在花中的表達(dá)量最高;且GhAREB4基因表達(dá)受到干旱、高鹽、低溫、脫落酸(ABA)等處理的誘導(dǎo),其可能調(diào)控棉花對(duì)非生物逆境的耐性響應(yīng)。研究結(jié)果為進(jìn)一步研究該基因?qū)γ藁湍嬲{(diào)控機(jī)制奠定了基礎(chǔ)。
[Abstract]:AREBs transcription factor family genes are mainly involved in drought, high salt, low temperature stress response, and play a very important role in plant resistance to various stresses. In this study, GhAREB4 gene of Upland cotton was cloned by sequence electron splicing. The length of the gene was 1784 BP, the open reading frame was 1 227 BP, encoding 408 amino acids, the predicted molecular weight was 44.3 KD and the isoelectric point was 8.88. It was found that the protein secondary structure contained the conserved domain of bZIP gene family. Phylogenetic tree analysis showed that GhAREB4 and cocoa had the highest homology with AREB transcription factors. The subcellular localization analysis of green fluorescent protein showed that the expression of GhAREB4 gene was the highest in flower, and the expression of GhAREB4 gene was induced by drought, high salt, low temperature, abscisic acid (ABA), etc. It may regulate cotton tolerance to abiotic stress. The results laid a foundation for further study of the stress tolerance regulation mechanism of the gene.
