腹腔注射亞精胺對(duì)小鼠卵巢組織多胺含量及代謝相關(guān)基因表達(dá)的影響

【摘要】:【目的】研究腹腔注射亞精胺對(duì)鼠卵巢組織多胺代謝的影響!痉椒ā拷o鼠腹腔注射不同劑量亞精胺[0(對(duì)照組、0.05、0.10和0.15 mg·g~(-1)],應(yīng)用實(shí)時(shí)熒光定量PCR檢測(cè)多胺代謝關(guān)鍵基因表達(dá)量,應(yīng)用高效液相色譜檢測(cè)鼠卵巢組織中多胺含量!窘Y(jié)果】注射0.15 mg·g~(-1)亞精胺時(shí),卵巢組織ODC、OAZ1、SPMS、SSAT、PAOX和SMOX基因表達(dá)量顯著高于其他3組;注射亞精胺組鼠卵巢組織SPDS表達(dá)量均顯著低于對(duì)照組;注射0.10 mg·g-1亞精胺組鼠卵巢組織中腐胺和亞精胺含量均顯著高于對(duì)照組,而注射亞精胺對(duì)卵巢中精胺含量無(wú)顯著影響!窘Y(jié)論】外源性亞精胺的腹腔注射可導(dǎo)致鼠卵巢組織中多胺含量以及多胺代謝相關(guān)基因表達(dá)量發(fā)生變化,且具有劑量依賴性,提示外源性亞精胺可通過(guò)介導(dǎo)卵巢組織的多胺代謝來(lái)參與調(diào)控卵巢功能。
[Abstract]:[objective] to study the effect of intraperitoneal injection of spermidine on the metabolism of polyamines in ovarian tissues of rats. [methods] different doses of spermidine were injected intraperitoneally into mice [0 (control group: 0.050.10 and 0.15 mg g ~ (-1)], and real-time quantitative PCR was used to detect the expression of key genes in polyamine metabolism. High performance liquid chromatography (HPLC) was used to detect the content of polyamines in mouse ovary. [results] the expression of ODCOAZ1 SPMSMSSSATPAOX and SMOX genes in ovarian tissue was significantly higher than that in the other three groups after 0.15 mg g ~ (-1) spermidine injection. The expression of SPDS in ovarian tissue of rats injected with spermidine was significantly lower than that of control group, and the contents of putrescine and spermidine in ovarian tissue of rats injected with 0.10 mg / g spermine were significantly higher than those in control group. There was no significant effect of spermidine injection on the content of spermine in ovary. [conclusion] Intraperitoneal injection of exogenous spermidine can result in changes of polyamine content and the expression of polyamine metabolism-related genes in ovarian tissue in a dose-dependent manner. The results suggest that exogenous spermidine can regulate ovarian function by mediating the metabolism of polyamines in ovarian tissues.
