【摘要】：實(shí)驗(yàn)克隆了興國(guó)紅鯉(Cyprinus carpio var singuonensis)雄激素受體(androgen receptor,AR)基因的全長(zhǎng)c DNA序列和卵黃蛋白原(vitellogenin,vtg)基因的部分c DNA序列,并對(duì)雌性個(gè)體組織及甲基睪丸酮(Methyltestosterone,MT)暴露下幼魚肝胰腺(以下簡(jiǎn)稱:肝)中AR和vtg的表達(dá)進(jìn)行檢測(cè)。興國(guó)紅鯉AR基因c DNA全長(zhǎng)3 229 bp,包括104 bp的5'非編碼區(qū)(untranslation region,UTR)、編碼846個(gè)氨基酸的2 541 bp開放閱讀框(open reading frame,ORF)和485 bp 3'UTR。氨基酸序列同源性分析表明,興國(guó)紅鯉AR與其他鯉科魚類AR的同源性較高。組織表達(dá)特征研究表明,AR在雌性個(gè)體的肝、卵巢、中腎、肌肉和端腦中有表達(dá),其中肝中的表達(dá)量最高;vtg主要在肝、端腦和鰓中表達(dá)。興國(guó)紅鯉幼魚在50μg/L的MT中暴露4周后,采用qRT-PCR檢測(cè)了肝中AR和vtg的表達(dá)情況。在處理第1、2周,AR的表達(dá)受到一定的抑制,而在第3、4周其表達(dá)量升高,但其表達(dá)無(wú)顯著性變化;而vtg的表達(dá)量在第3、4周均有極顯著的升高。
[Abstract]:The full-length c DNA sequence of the androgen receptor AR gene and the partial c DNA sequence of the vitellogenin VTG gene of Cyprinus carpio var singuonensis) were cloned. The expression of AR and vtg in the hepatopancreas of juvenile fish (hereinafter referred to as liver) was detected in female individual tissues and methyltestosterone (MT). The total length of AR gene of Xingguo Red Carp is 3 229 BP, including 104bp untranslation region UTR, 2 541bp open reading frame (open reading frame ORF) encoding 846 amino acids, and 485 BP 3G UTR. The amino acid sequence homology analysis showed that the AR of Xingguo red carp had high homology with that of other Cyprinidae fishes. The study of tissue expression showed that the expression of AR was found in the liver, ovary, mesonephrosis, muscle and telencephalon of female individuals, and the highest expression of VTG was found in liver, telencephalon and Gill. After exposed to 50 渭 g / L MT for 4 weeks, the expression of AR and vtg in liver was detected by qRT-PCR. The expression of AR was inhibited at the 1st week, but increased at the 3rd week, but the expression of vtg increased significantly at the 3th week, while the expression of vtg increased significantly at the 4th week.
