導(dǎo)入iaaM與ipt雙價(jià)基因的轉(zhuǎn)基因大豆檢測(cè)及表達(dá)效果研究

【摘要】:大豆是我國的重要的油料作物和經(jīng)濟(jì)作物,但其單產(chǎn)卻遠(yuǎn)遠(yuǎn)低于歐美國家,因此提高其單產(chǎn)育種對(duì)國民的生產(chǎn)實(shí)踐具有重要的意義。主要通過GUS組織化學(xué)染色分析、PCR檢測(cè)、Southern blot檢測(cè)技術(shù)對(duì)轉(zhuǎn)iaa M與ipt雙價(jià)基因的T1代轉(zhuǎn)基因大豆進(jìn)行檢測(cè),結(jié)果表明有5個(gè)株系iaa M和ipt基因以單拷貝的形式整合進(jìn)了大豆基因組。采用半定量RT-PCR檢測(cè)表明有4個(gè)株系外源雙價(jià)基因高效表達(dá)。最后經(jīng)產(chǎn)量性狀考種結(jié)果表明:有3個(gè)株系的表現(xiàn)較好,其中株系4增產(chǎn)12.85%。
[Abstract]:Soybean is an important oil crop and cash crop in China, but its yield per unit is much lower than that in Europe and America. Gus histochemical staining was used to detect T 1 transgenic soybean with iaa M and ipt bivalent genes. The results showed that 5 lines iaa M and ipt genes were integrated into soybean genome in single copy form. Semi-quantitative RT-PCR analysis showed that there were 4 lines with high expression of exogenous bivalent genes. At last, the results of yield test showed that three lines had better performance, among them, line 4 increased the yield by 12.85kum.
