導入iaaM與ipt雙價基因的轉基因大豆檢測及表達效果研究

【摘要】:大豆是我國的重要的油料作物和經濟作物,但其單產卻遠遠低于歐美國家,因此提高其單產育種對國民的生產實踐具有重要的意義。主要通過GUS組織化學染色分析、PCR檢測、Southern blot檢測技術對轉iaa M與ipt雙價基因的T1代轉基因大豆進行檢測,結果表明有5個株系iaa M和ipt基因以單拷貝的形式整合進了大豆基因組。采用半定量RT-PCR檢測表明有4個株系外源雙價基因高效表達。最后經產量性狀考種結果表明:有3個株系的表現(xiàn)較好,其中株系4增產12.85%。
[Abstract]:Soybean is an important oil crop and cash crop in China, but its yield per unit is much lower than that in Europe and America. Gus histochemical staining was used to detect T 1 transgenic soybean with iaa M and ipt bivalent genes. The results showed that 5 lines iaa M and ipt genes were integrated into soybean genome in single copy form. Semi-quantitative RT-PCR analysis showed that there were 4 lines with high expression of exogenous bivalent genes. At last, the results of yield test showed that three lines had better performance, among them, line 4 increased the yield by 12.85kum.
