基因間插入對(duì)水稻農(nóng)藝性狀及刺吸式口器害蟲取食選擇性與種群密度的影響

【摘要】:本文評(píng)價(jià)了T-DNA插入對(duì)水稻農(nóng)藝性狀與抗蟲性、褐飛虱Nilaparvata lugens St?l取食選擇性及3種刺吸式口器害蟲種群密度的影響,以分析轉(zhuǎn)基因操作及外源基因插入可能引起的非預(yù)期效應(yīng)。結(jié)果表明,供試ZD1640-3、ZD1640-19、ZD1640-23和ZD1640-24共4個(gè)突變體株系T-DNA插入位點(diǎn)附近基因的表達(dá)均不受影響;除個(gè)別株系的株高、每穗粒數(shù)或結(jié)實(shí)率有降低外,主要農(nóng)藝性狀參數(shù)與受體親本無顯著差異。除ZD1640-24外,突變體各株系上褐飛虱、白背飛虱Sogatella furcifera(Horváth)和黑尾葉蟬Nephotettix bipunctatus(Fabricius)平均種群密度與受體親本無顯著差異;且4個(gè)突變體株系上3種害蟲種群密度在大多數(shù)情況下與受體親本也無顯著差異。與受體親本相比,突變體株系ZD1640-24對(duì)褐飛虱的抗性及取食選擇性無顯著影響。綜合分析認(rèn)為,T-DNA基因間插入并沒有引起水稻發(fā)生顯著的非預(yù)期變化,個(gè)別突變體某些考查指標(biāo)的變化主要來自于個(gè)體差異。
[Abstract]:The effects of T-DNA insertion on agronomic traits and insect resistance of rice, feeding selectivity of Nilaparvata lugens Stl and population density of three kinds of sucking insect pests were evaluated in order to analyze the unexpected effects caused by transgenic operation and exogenous gene insertion. The results showed that the expression of genes near the T-DNA insertion site of ZD1640-3 and ZD1640-24 mutant lines were not affected, except for the plant height, grain number per panicle or seed setting rate of individual lines, there was no significant difference between the main agronomic characters and the recipient parents. Except for ZD1640-24, there was no significant difference between the average population density of BPH, Sogatella furcifera (Horv 謾 th) and Nephotettix bipunctatus (Fabricius). The population densities of the three pests in the four mutants were not significantly different from those of the recipient parents in most cases. Compared with the recipient parents, the mutant ZD1640-24 had no significant effect on the resistance and feeding selectivity of brown planthopper. The comprehensive analysis showed that the insertion of T-DNA gene did not cause significant unexpected changes in rice, and the changes of some indexes in individual mutants were mainly due to individual differences.
