通關(guān)藤提取物誘導(dǎo)BGC-823細(xì)胞凋亡及對(duì)MTDH基因表達(dá)的作用研究

【摘要】:目的:通過觀察不同濃度通關(guān)藤提取物對(duì)人胃腺癌BGC-823細(xì)胞增殖、凋亡及MTDH基因表達(dá)的影響,為胃癌的臨床治療提供實(shí)驗(yàn)基礎(chǔ)及潛在治療靶點(diǎn)。方法:(1)MTT法檢測(cè)通關(guān)藤提取物對(duì)BGC-823細(xì)胞增殖影響。(2)Annexin V/PI雙染法檢測(cè)通關(guān)藤提取物對(duì)BGC-823細(xì)胞凋亡影響。(3)反轉(zhuǎn)錄多聚酶鏈反應(yīng)(RTPCR)檢測(cè)通關(guān)藤提取物對(duì)BGC-823細(xì)胞MTDH mRNA表達(dá)影響。(4)Western Blot法檢測(cè)通關(guān)藤提取物對(duì)BGC-823細(xì)胞凋亡相關(guān)蛋白表達(dá)影響。結(jié)果:(1)通關(guān)藤提取物作用于BGC-823細(xì)胞,細(xì)胞生長(zhǎng)抑制率隨藥物濃度增大而升高,加藥各組與對(duì)照組相比,差異有統(tǒng)計(jì)學(xué)意義。(2)通關(guān)藤提取物0 mmol/L、0.5 mmol/L、1 mmol/L、2mmol/L誘導(dǎo)BGC-823細(xì)胞凋亡率為(2.98±1.01)%、(6.79±0.68)、(12.512±1.27)%、(16.97±1.09),與對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義。(3)通關(guān)藤提取物可下調(diào)BGC-823細(xì)胞MTDH mRNA表達(dá),這種作用具有濃度依賴性。(4)通關(guān)藤提取物能夠下調(diào)Bcl-2家族中的抑凋亡因子Bcl-2、Bcl-xl的表達(dá)尤其是Bcl-xl的表達(dá),上調(diào)凋亡執(zhí)行者Caspase-3的表達(dá)。結(jié)論:通關(guān)藤提取物可下調(diào)BGC-823細(xì)胞MTDH mRNA表達(dá),并通過下調(diào)Bcl-2、Bcl-xl,上調(diào)Caspase-3的表達(dá)而誘導(dǎo)BGC-823細(xì)胞凋亡,從而抑制了BGC-823細(xì)胞的生長(zhǎng)增殖。
[Abstract]:Aim: to investigate the effects of different concentrations of Tungkuan extract on the proliferation, apoptosis and MTDH gene expression of human gastric adenocarcinoma cell line BGC-823 in order to provide experimental basis and potential therapeutic target for the clinical treatment of gastric cancer. Methods MTT assay was used to detect the proliferation of BGC-823 cells. The effect of Annexin V / Pi double staining on the apoptosis of BGC-823 cells was detected by MTT) reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of MTDH mRNA in BGC-823 cells by reverse transcription polymerase chain reaction (RT-PCR). The expression of apoptosis-related protein in BGC-823 cells was detected by Western blot. Results the growth inhibition rate of BGC-823 cells increased with the increase of drug concentration. The apoptosis rate of BGC-823 cells induced by 0 mmol / L 0 mmol / L ~ (0.5) mmol / L ~ (1) mmol / L ~ (2) / L was 2.98 鹵1.01% (P < 0.05). The expression of MTDH mRNA in BGC-823 cells was significantly lower than that in the control group (P < 0.05), and the expression of MTDH mRNA in BGC-823 cells was significantly lower than that in the control group (P < 0.05), and the expression of MTDH mRNA in BGC-823 cells was significantly lower than that in the control group (P < 0.05), and the expression of MTDH mRNA in BGC-823 cells was significantly lower than that in control group. This effect was concentration-dependent. The extract of Tongguan vine could down-regulate the expression of Bcl-2 family inhibitor Bcl-2nBcl-xl, especially the expression of Bcl-xl, and up-regulate the expression of Caspase-3 in apoptotic executors. Conclusion: the extract of Tongguan vine can down-regulate the expression of MTDH mRNA in BGC-823 cells and inhibit the growth and proliferation of BGC-823 cells by down-regulating the expression of Bcl-2OBcl-xland up-regulating the expression of Caspase-3 in BGC-823 cells.
