棉花中HD-Zip Ⅳ家族基因的全基因組鑒定及特征分析

【摘要】:同源異型亮氨酸拉鏈蛋白(homeodomain-leucine zipper,HD-Zip)是高等植物特有的一類轉(zhuǎn)錄因子,分成Ⅰ~Ⅳ4個(gè)亞家族。HD-ZipⅣ轉(zhuǎn)錄因子主要參與表皮結(jié)構(gòu)的發(fā)育調(diào)控,在其他作物中已經(jīng)廣泛研究,但是棉花中還未進(jìn)行系統(tǒng)的分析。本研究中,我們分別對(duì)陸地棉、亞洲棉、雷蒙德氏棉進(jìn)行全基因組的鑒定,分別得到15個(gè)、12個(gè)、14個(gè)HD-ZipⅣ基因。除一個(gè)雷蒙德氏棉基因發(fā)生丟失外,陸地棉中幾乎保留了所有的亞洲棉、雷蒙德氏棉中的HD-ZipⅣ基因。分析表明這些基因編碼的蛋白都有HD-ZipⅣ轉(zhuǎn)錄因子特征性的結(jié)構(gòu)域,且進(jìn)化樹(shù)上關(guān)系近的成員擁有相似的基因結(jié)構(gòu)和基序組成。大部分陸地棉HD-ZipⅣ基因經(jīng)歷了純化選擇,且有一些經(jīng)歷正向選擇。同時(shí),進(jìn)一步分析得出結(jié)論:片段復(fù)制在陸地棉HD-ZipⅣ基因的演化中起重要作用。組織表達(dá)分析表明大部分HD-ZipⅣ基因主要在發(fā)育著的胚珠和纖維中表達(dá),暗示該家族與棉花中種子形成、纖維發(fā)育有關(guān)?傊,本研究為棉花中HD-ZipⅣ基因的功能研究及應(yīng)用提供了參考依據(jù)。
[Abstract]:Homologous homeodomain-leucine zipper (HD-Zip) is a specific transcription factor of higher plants. It is divided into 4 subfamilies, I ~ IV,.HD-Zip IV transcription factors, which are mainly involved in the development and regulation of epidermal structure. It has been widely studied in other crops, but it has not been systematically analyzed in cotton. In this study, we 15, 12, and 14 HD-Zip IV genes were identified for land cotton, Asian cotton and Raymond's cotton respectively. In addition to the loss of a Raymond's cotton gene, almost all of the Asian cotton and the HD-Zip IV gene in Raymond S cotton were retained in the land cotton. The analysis showed that all the genes encoded by these genes were HD-Zip (IV) the characteristic domain of the transcription factor, and the members of the evolutionary tree have similar genetic structure and motif. Most of the HD-Zip IV gene of upland cotton has been selected and has some positive selection. Tissue expression analysis showed that most of HD-Zip IV genes were expressed mainly in the developed ovules and fibers, suggesting that the family was related to the formation of seeds and fiber development in cotton. In a word, this study provides a reference for the function research and application of HD-Zip IV gene in cotton.
