轉基因油菜Ms8質(zhì)粒分子的制備

【摘要】:隨著轉基因產(chǎn)品的大規(guī)模商業(yè)化和我國大規(guī)模進口油菜籽,有效管控存在的風險顯得非常必要和有意義,試驗研究了質(zhì)粒標準分子pMs8的研制過程,包括質(zhì)粒分子的構建、純度分析、特異性檢測、均勻性檢驗、穩(wěn)定性考察、定值。結果表明,該質(zhì)粒分子標準物質(zhì)的內(nèi)源基因和外源基因的均勻性良好,沒有擴增出所選的其他特異性基因,短期穩(wěn)定性在14 d(處理溫度大于25℃)發(fā)生顯著變化,長期穩(wěn)定性大于6個月,兩種方法對外源基因拷貝數(shù)的定值結果相近,因此,p Ms8是有準確量值的質(zhì)粒標準分子?商娲匝芯拷Y果表明,其可替代基因組作為轉基因油菜Ms8定量的陽性標準。
[Abstract]:With the large-scale commercialization of transgenic products and the large-scale import of rapeseed in China, it is necessary and meaningful to effectively control the risks. The development process of plasmid standard molecule pMs8, including the construction of plasmid molecule, is studied experimentally. Purity analysis, specificity test, homogeneity test, stability test, fixed value.
