谷氨酸棒桿菌ATCC14067海藻糖合酶基因的克隆及功能鑒定

【摘要】:海藻糖分子是由2個(gè)吡喃葡萄糖環(huán)以α,α-1,1-糖苷鍵連接而成的一種十分穩(wěn)定的非還原二糖,由于其特殊的生物學(xué)保護(hù)功用,廣泛應(yīng)用于醫(yī)藥、化妝品、食品等多個(gè)領(lǐng)域,具有很好的市場前景。目前,海藻糖的生產(chǎn)方法以生物酶法制備為主。其中,利用Tre Y-Tre Z途徑由淀粉生產(chǎn)海藻糖和利用Tre S途徑由麥芽糖生產(chǎn)海藻糖最為經(jīng)濟(jì)可行。針對(duì)谷氨酸棒桿菌ATCC14067的海藻糖合酶基因(tre S)進(jìn)行基因克隆,將其插入表達(dá)質(zhì)粒p ET-22b,在宿主菌E.coli BL21(DE3)中進(jìn)行誘導(dǎo)表達(dá),成功得到特異表達(dá)的目的蛋白。經(jīng)高效液相色譜檢測證實(shí),該重組酶作用于麥芽糖后,可以產(chǎn)生海藻糖,具有海藻糖合酶的相應(yīng)活性。在最適反應(yīng)條件(30℃,p H7.5)下海藻糖合酶的轉(zhuǎn)化效率可達(dá)60%~70%。實(shí)驗(yàn)系統(tǒng)地對(duì)谷氨酸棒桿菌ATCC14067的海藻糖合酶基因進(jìn)行了異源表達(dá)及酶學(xué)性質(zhì)的研究,為其大規(guī)模生產(chǎn)及工業(yè)化應(yīng)用奠定了基礎(chǔ)。
[Abstract]:Trehalose molecule is a kind of stable non-reducing disaccharide which is composed of two glucopyranosaccharide rings connected by 偽, 偽 -1ta-1-glucoside bond. Due to its special biological protective function, trehalose is widely used in many fields, such as medicine, cosmetics, food, etc. There is a good market prospect. At present, trehalose is mainly produced by biological enzyme method. Among them, trehalose production from starch by Tre Y-Tre Z pathway and trehalose from maltose by Tre S pathway is the most economical and feasible. The trehalose synthase gene of Corynebacterium glutamicum ATCC14067 was cloned and inserted into the expression plasmid pET-22b. High performance liquid chromatography (HPLC) showed that the recombinant enzyme could produce trehalose and had the corresponding activity of trehalose synthase after acting on maltose.
