【摘要】：目的探討DNA甲基化老年大鼠學(xué)習(xí)記憶相關(guān)基因egr1表達(dá)變化的機(jī)制及補(bǔ)腎益氣方藥——左歸丸、益氣聰明湯的調(diào)整作用。方法以自然衰老(24月齡)SD雄性大鼠為動物模型,隨機(jī)分為老年對照組、老年補(bǔ)腎組、老年益氣組、老年合劑組和老年拮抗組,另設(shè)一組青年對照組(5月齡)。采用RT-PCR法及Western印跡法觀察大鼠學(xué)習(xí)記憶相關(guān)基因egr1 mRNA和蛋白表達(dá)變化,采用亞硫酸氫鹽轉(zhuǎn)化結(jié)合高通量測序法檢測基因egr1啟動子區(qū)甲基化狀態(tài),同時觀察補(bǔ)腎益氣方藥對上述指標(biāo)變化的調(diào)整作用。結(jié)果與青年對照組相比,老年大鼠egr1基因mRNA和蛋白表達(dá)均明顯降低(P0.05),其啟動子區(qū)甲基化增高,左歸丸、益氣聰明湯能不同程度地改善egr1的表達(dá),并降低egr1啟動子區(qū)甲基化率。結(jié)論補(bǔ)腎益氣方藥改善老年大鼠增齡性學(xué)習(xí)記憶退化可能與抑制基因egr1啟動子DNA甲基化而提高egr1的表達(dá)有關(guān)。
[Abstract]:Objective to investigate the mechanism of egr1 expression of learning and memory related genes in aged rats with methylation and the regulating effect of Zuo Gui Pill, a prescription of tonifying kidney and tonifying qi. Methods SD male rats aged 24 months were randomly divided into old control group, senile tonifying kidney group, aged invigorating qi group, aged mixture group and aged antagonist group, and a young control group was set up. RT-PCR and Western blot were used to observe the expression of learning and memory related gene egr1 and protein in rats. The methylation status of gene egr1 promoter was detected by bisulfite transformation and high throughput sequencing. At the same time, observe the effect of tonifying kidney and tonifying qi prescription on the change of above indexes. Results compared with the young control group, the expression of egr1 gene and protein in the aged rats were significantly lower than those in the young control group. The methylation of the promoter region of the aged rats was increased. Zuo Gui Pill and Yiqizeng decoction could improve the expression of egr1 and decrease the methylation rate of the egr1 promoter in varying degrees. Conclusion the effect of Bushen Yiqi recipe on aging learning and memory degeneration in aged rats may be related to the increase of egr1 expression by methylation of egr1 promoter.
