【摘要】：實(shí)時(shí)熒光定量PCR(qRT-PCR)是研究基因表達(dá)的重要手段之一,而選擇合適的內(nèi)參基因是獲得可靠研究結(jié)果的基礎(chǔ).本研究以世界性分布的重要倉(cāng)儲(chǔ)害蟲(chóng)印度谷螟(Plodia interpunctella)為研究對(duì)象,旨在篩選出該蟲(chóng)不同發(fā)育階段及不同品系qRT-PCR的最適內(nèi)參基因,并利用兩個(gè)獨(dú)立軟件(ge Norm和Norm Finder)對(duì)8個(gè)備選內(nèi)參基因的表達(dá)穩(wěn)定性進(jìn)行了評(píng)價(jià).結(jié)果表明,不同發(fā)育階段最穩(wěn)定表達(dá)的內(nèi)參基因?yàn)镋f1a,而不同品系最穩(wěn)定的是b-Actin和Ef1a.利用篩選到的Ef1a,b-Actin及兩者組合對(duì)目標(biāo)基因(過(guò)氧化物酶POD)進(jìn)行標(biāo)準(zhǔn)化,印度谷螟不同品系間的POD基因表達(dá)量存在顯著差異,而以穩(wěn)定性最差的SD作為內(nèi)參時(shí),不同品系間POD的相對(duì)表達(dá)量并無(wú)顯著差異.這表明,在開(kāi)展qRT-PCR研究時(shí)評(píng)價(jià)內(nèi)參基因的穩(wěn)定性非常必要,且基于篩選到的最佳基因作為內(nèi)參時(shí),才能準(zhǔn)確獲得目標(biāo)基因的相對(duì)表達(dá)水平.這些結(jié)果將有助于更好地研究印度谷螟發(fā)育及品系特異相關(guān)基因的功能,并為篩選印度谷螟在其他條件下的最適內(nèi)參基因提供參考.
[Abstract]:Real-time quantitative PCR qRT-PCR is one of the important methods to study gene expression, and the selection of suitable internal reference gene is the basis for obtaining reliable results. In this study, Plodia interpunctura, a worldwide important storage pest, was used to screen the optimal internal reference genes for different stages of development and different strains of qRT-PCR. The stability of the expression of 8 candidate reference genes was evaluated by using two independent softwares named ge Norm and Norm Finder. The results showed that the most stable endoparameters expressed in different developmental stages were Ef1a, while the most stable ones in different strains were b-Actin and Ef1a. The target gene (POD) was standardized by using the selected Ef1aPb-Actin and their combinations. There were significant differences in the expression of POD gene among different strains of Indian grain borer, but when SD with the lowest stability was used as the internal control. There was no significant difference in the relative expression of POD among different strains. This indicates that it is necessary to evaluate the stability of the internal reference gene in the qRT-PCR study, and the relative expression level of the target gene can be accurately obtained only when the best selected gene is used as the internal reference. These results will be helpful to study the development of Indian grain borer and the function of strain specific genes, and provide a reference for screening the most suitable internal reference genes of Indian grain borer under other conditions.
