玉米熱激轉(zhuǎn)錄因子基因ZmHsf25的克

【摘要】:植物熱激轉(zhuǎn)錄因子(heat shock transcription factor,Hsf)對下游熱激蛋白基因及耐熱性相關(guān)基因的表達起關(guān)鍵調(diào)控作用。玉米中至少有30個Hsf家族成員,其中B族有7個。在前期對玉米A1亞族熱激轉(zhuǎn)錄因子基因ZmHsf06克隆及特性、抗旱耐熱性功能分析的基礎(chǔ)上,從玉米幼葉中克隆到B族熱激轉(zhuǎn)錄因子基因ZmHsf25的完整編碼序列,序列全長957 bp,編碼318個氨基酸殘基。蛋白質(zhì)序列含有DNA結(jié)合結(jié)構(gòu)域、核定位信號序列和核輸出信號序列,同源分析結(jié)果顯示,ZmHsf25與高粱Sb06g025710的相似性最高,達92%。熒光定量分析表明,正常生長條件下,ZmHsf25在玉米多個組織器官中表達,幼嫩花粉中高表達,幼嫩根系和雌穗中表達量較低。42℃熱脅迫能顯著上調(diào)根系和葉片ZmHsf25的表達,葉片中上升幅度高于根系,處理50 min達到峰值;正常條件下,水楊酸和H2O2處理均使根系和葉片ZmHsf25表達下調(diào),而熱激條件下卻表現(xiàn)為顯著上調(diào)。通過在洋蔥表皮細(xì)胞中瞬時表達并觀察GFP熒光,確定ZmHsf25定位在細(xì)胞核。通過轉(zhuǎn)化酵母并進行熱脅迫處理發(fā)現(xiàn),熱脅迫同時降低正常和轉(zhuǎn)ZmHsf25酵母的耐熱性,但與轉(zhuǎn)空載體對照相比,轉(zhuǎn)基因酵母的耐熱性更強。推測ZmHsf25參與玉米熱脅迫響應(yīng)和花粉的發(fā)育過程,可能是水楊酸熱激信號轉(zhuǎn)導(dǎo)途徑的下游組分。研究結(jié)果為進一步探明玉米熱激轉(zhuǎn)錄因子家族成員的特性與功能提供了依據(jù)。
[Abstract]:Heat shock transcription factor-Hsf) plays a key role in regulating the expression of heat shock protein genes and heat tolerance related genes. There are at least 30 Hsf family members in maize, including 7 in B group. Based on the cloning and characterization of heat shock transcription factor gene (ZmHsf06) of subfamily A1 and the function analysis of drought resistance and heat tolerance, the complete coding sequence of group B heat shock transcription factor gene ZmHsf25 was cloned from young leaves of maize. The sequence was 957 BP, encoding 318 amino acid residues. The protein sequence contained DNA binding domain, nuclear localization signal sequence and nuclear output signal sequence. The homology analysis showed that ZmHsf25 had the highest similarity with sorghum Sb06g025710, and reached 92 points. Fluorescence quantitative analysis showed that ZmHsf25 was expressed in several tissues and organs of maize under normal growth conditions, high in young pollen, and significantly up-regulated ZmHsf25 expression in roots and leaves under low heat stress at 0.42 鈩,
