子癇前期宮內(nèi)環(huán)境導(dǎo)致子代大鼠海馬HPA軸相關(guān)基因表達改變

【摘要】:目的研究子癇前期宮內(nèi)環(huán)境對子代SD大鼠海馬下丘腦-垂體-腎上腺(hypothalamic-pituitaryadrenal,HPA)軸相關(guān)基因表達的影響。方法通過給妊娠第14天SD大鼠連續(xù)皮下注射亞硝基左旋精氨酸甲酯(L-NAME)至分娩的方法建立子癇前期大鼠動物模型,對照組予生理鹽水1 mL皮下注射。用酶聯(lián)免疫檢測(ELISA)方法檢測新生子代大鼠血皮質(zhì)酮(corticosterone,CORT)濃度。用Real-time PCR方法和Western bolt分別檢測8周齡子代大鼠海馬組織糖皮質(zhì)激素受體(glucocorticoid receptor,GR)、促腎上腺皮質(zhì)激素釋放激素(corticotropin releasing hormone,CRH)、CRH受體1(CRH receptor 1,CRHR1)和IL-6的mRNA水平及其蛋白質(zhì)水平。結(jié)果與正常子代新生鼠相比,子癇前期子代新生鼠血CORT濃度升高(P0.05)。與正常子代鼠相比,子癇前期青少年子代鼠海馬GR、CRH和CRHR1 mRNA和蛋白質(zhì)水平均增加(P0.05),炎性因子IL-6mRNA和蛋白質(zhì)水平均增加(P0.05)。結(jié)論子癇前期宮內(nèi)環(huán)境影響子代大鼠海馬HPA軸相關(guān)基因的表達,在子代學習記憶能力受損中起作用。
[Abstract]:Objective to study the effect of intrauterine environment of preeclampsia on the expression of genes related to the axis of hypothalamic-pituitary-adrenal gland (HPAA) in rat hippocampus. Methods the preeclampsia rat model was established by subcutaneous injection of L-NAME-nitroso-L-arginine methyl ester into labor on the 14th day of pregnancy, and the control group was subcutaneously injected with 1 mL normal saline. The plasma corticosterone (Cort) concentration in newborn rats was detected by Elisa. The levels of glucocorticoid receptor (Glucocorticoid receptor), corticotropin releasing hormone (corticotropin releasing CRH receptor 1(CRH receptor 1CRHR1) and IL-6 and their protein levels in hippocampus of 8-week-old rats were detected by Real-time PCR and Western bolt, respectively. Results compared with normal newborn mice, the serum CORT concentration of preeclampsia newborn rats was higher than that of normal offspring. Compared with normal offspring, the levels of CRH, CRHR1 mRNA and protein in hippocampal GRG of juvenile preeclampsia rats were increased, while the levels of IL-6mRNA and protein of inflammatory factors were increased. Conclusion the intrauterine environment of preeclampsia affects the expression of HPA axis related genes in the hippocampus of the offspring and plays an important role in the impairment of learning and memory ability of the offspring.
