【摘要】：生物信息學(xué)分析發(fā)現(xiàn)西(甜)瓜果斑病菌(Acidovorax citrulli,Ac)基因組中存在水稻條斑病菌(Xanthomonas oryzae pv.oryzicola,Xoc)avrRxo1的同源基因avrRxo1_(Ac)。為了揭示avrRxo1_(Ac)是否能夠激發(fā)玉米R(shí)基因Rxo1介導(dǎo)的過(guò)敏反應(yīng)(Hypersensitive response,HR),本研究克隆了avrRxo1_(Ac)基因;氨基酸同源性分析結(jié)果顯示:AvrRxo1_(Ac)與AvrRxo1的同源性為53.16%。將avrRxo1_(Ac)置于Xoc的avrRxo1突變體中,發(fā)現(xiàn)其不能恢復(fù)該突變體在含有Rxo1玉米上激發(fā)HR反應(yīng)的能力;借助農(nóng)桿菌瞬時(shí)表達(dá)體系發(fā)現(xiàn)AvrRxo1A,也不能在Rxo1玉米上激發(fā)HR反應(yīng)。這些結(jié)果表明avrRxo1_(Ac)-Rxo1互作不表現(xiàn)為基因-對(duì)-基因關(guān)系,暗示Rxo1抗病基因轉(zhuǎn)入西甜瓜中,將不能起到抗西(甜)瓜細(xì)菌性果斑病的作用。
[Abstract]:Bioinformatics analysis showed that Xanthomonas oryzae pv.oryzicola1, a homologous gene of Acidovorax citrullilio1, was found in the genome of Acidovorax citrulliliae (Acidovorax citrullifolia), and the homologous gene, avrRxo1, was found in Acidovorax citrullii Ac. In order to find out whether avrRxo1ac) can stimulate the allergic response mediated by Rxo1 of R gene in maize, we cloned the AvrRxo1C) gene, and the result of amino acid homology analysis showed that the homology of AvrRxo1 was 53.16%. It was found that AvrRxo1A was not able to restore the ability of the mutant to stimulate HR response on Rxo1 containing maize, and that AvrRxo1A could not stimulate HR response in Rxo1 maize with the help of Agrobacterium tumefaciens transient expression system. These results indicated that the interaction of avrRxo1_(Ac)-Rxo1 did not show the relationship between genes and genes, suggesting that Rxo1 resistance genes transferred into western muskmelon would not play the role of resistance to bacterial fruit spot disease in western (sweet) melon.
