污泥臭氧氧化處理過程中活菌抗藥基因豐度的消減

【摘要】:臭氧氧化技術(shù)是一種廣泛應(yīng)用的污泥減量技術(shù),然而臭氧處理能否對污泥中的抗生素抗藥基因進(jìn)行有效消減還不清楚。采用單疊氮溴化丙錠(propidium monoazide,PMA)預(yù)處理結(jié)合定量PCR(qPCR)方法對污泥臭氧減量過程中不同臭氧消耗量下活性污泥活菌中的四環(huán)素、氨基糖苷和大環(huán)內(nèi)酯3大類共21種抗藥基因的變化進(jìn)行了研究。結(jié)果表明:臭氧氧化可以有效消減活性污泥活菌中21種抗藥基因的絕對豐度(每毫升污泥的抗藥基因拷貝數(shù)),在臭氧消耗量0.31 g·g~(-1)(TSS)情況下抗藥基因總量降低了75.44%;盡管絕對豐度下降,污泥臭氧氧化處理過程中大量抗藥基因的相對豐度(抗藥基因拷貝數(shù)與細(xì)菌16S rRNA基因拷貝數(shù)的比例)逐漸增加,表明臭氧處理后污泥中耐藥菌占總活菌比例可能增加,具有一定的抗性傳播風(fēng)險;Ⅰ型整合子是抗藥基因水平轉(zhuǎn)移的重要遺傳元件,其絕對豐度同樣隨著臭氧消耗量增加而降低,而相對豐度逐漸上升。
[Abstract]:Ozone oxidation is a widely used sludge reduction technology. However, it is not clear whether ozone treatment can effectively reduce antibiotic resistance genes in sludge. The tetracycline in activated sludge living bacteria was treated with propidium monoazide PMA pretreatment combined with quantitative PCRX qPCRduring the process of sludge ozone reduction. The changes of 21 resistant genes of aminoglycoside and macrolide were studied. The results showed that ozone oxidation could effectively reduce the absolute abundance of 21 resistant genes (copy number of drug resistance genes per milliliter of sludge), and the total amount of drug resistant genes decreased by 75.44% under ozone consumption of 0.31 g / ml TSSs. The absolute abundance of the tube drops, The relative abundance of a large number of drug-resistant genes (the ratio of copy number of drug resistance gene to 16s rRNA gene) gradually increased during ozonation treatment of sludge, indicating that the proportion of drug-resistant bacteria in total live bacteria may increase after ozone treatment. Type I integron is an important genetic element for horizontal transfer of drug resistance genes, and its absolute abundance also decreases with the increase of ozone consumption, while the relative abundance increases gradually.
