香蕉花芽分化期抗枯萎病相關基因表達分析

【摘要】:通過對‘巴西蕉’和‘寶島蕉’花芽分化期響應枯萎病菌4號生理小種(Fusarium oxysporum f.sp.cubense race 4,Foc4)侵染的轉錄組進行分析,獲得了大量差異表達基因。本研究以‘巴西蕉’和‘寶島蕉’花芽分化期根部組織為材料,采用實時熒光定量PCR(q PCR)方法測定了10個與特定抗病機制相關基因的表達變化。結果表明,10個基因在‘巴西蕉’和‘寶島蕉’花芽分化過程中均有表達,但其表達量存在差異。與對照‘巴西蕉’相比,除laccase-4(A)基因在‘寶島蕉’中的表達量明顯下調外,laccase-4(B)、periplasmic beta-glucosidase precursor、glutathione-S-transferase Cla47、class Ⅲ peroxidase 136 precursor、peroxidase N、similar to T-phylloplanin、snakin-1、putative chitinase及cellulose synthase catalytic subunit 12等9個基因的表達量均上調,其中peroxidase N基因的表達量呈極顯著上調,相對表達量約為‘巴西蕉’的1 682倍。10個差異表達基因可能參與‘寶島蕉’花芽分化期對病原菌的防衛(wèi)反應,為進一步探討‘寶島蕉‘復雜的抗病防御反應分子機理奠定基礎。
[Abstract]:A large number of differentially expressed genes were obtained by analyzing the transcriptome of Fusarium oxysporum f.sp.cubense race 4 (Foc4), a physiological species of Fusarium oxysporum f.sp.cubense race 4, in response to the floral bud differentiation of 'Canna brasiliensis' and 'Baodao banana'. In this study, the expression changes of 10 genes related to specific disease resistance mechanism were determined by real-time fluorescence quantitative PCR(q method using the root tissue of 'Canna bassiana' and 'Baodao banana' during flower bud differentiation. The results showed that all of the 10 genes were expressed in the process of flower bud differentiation, but there were differences in their expression levels. Compared with the control, the expression of laccase-4) gene was significantly down-regulated in 'Baodao banana', except that the expression of laccase-4 beta-glucosidase precursorsortium-glutathione-S-transferase Cla47 class 鈪,
