水母雪蓮表皮毛發(fā)育相關(guān)基因SmTTG1的遺傳轉(zhuǎn)化研究

【摘要】:水母雪蓮(Saussurea medusa Maxim),又稱水母雪兔子,屬菊科風(fēng)毛菊屬,是一種生長(zhǎng)于青藏高原地區(qū)的珍稀、瀕危藥用植物。同時(shí),它又是一種典型的高山植物。植株密被表皮毛是水母雪蓮最具有代表性的形態(tài)特征,它是水母雪蓮對(duì)極端環(huán)境的一種適應(yīng)結(jié)構(gòu)。研究水母雪蓮表皮毛的形成和發(fā)育過(guò)程,對(duì)揭示高山植物適應(yīng)環(huán)境的特殊機(jī)制將具有重要的理論意義。為此,本研究以水母雪蓮作為實(shí)驗(yàn)材料,以表皮毛發(fā)育相關(guān)基因SmTTG1為研究對(duì)象,構(gòu)建了SmTTG1基因的植物表達(dá)載體,通過(guò)根癌農(nóng)桿菌為介導(dǎo),轉(zhuǎn)入普通煙草中。主要研究結(jié)果如下:(1)根據(jù)本實(shí)驗(yàn)室已克隆得到的水母雪蓮SmTTG1基因序列(Genbank登陸號(hào):KX447632),在其CDS兩側(cè)分別設(shè)計(jì)上下游引物,在上游引物5′端加入酶切位點(diǎn)SalⅠ,在下游引物5′端加入酶切位點(diǎn)Bam HⅠ,克隆了水母雪蓮SmTTG1基因的CDS序列(含酶切位點(diǎn))。(2)通過(guò)酶切法,將目的基因SmTTG1插入質(zhì)粒pRI 101-AN DNA的多克隆位點(diǎn),構(gòu)建了植物表達(dá)載體pRI 101-AN-SmTTG1,并通過(guò)凍融法將其導(dǎo)入農(nóng)桿菌C58C1中。(3)在根癌農(nóng)桿菌介導(dǎo)下,通過(guò)葉盤法將目的基因SmTTG1轉(zhuǎn)入普通煙草中。通過(guò)PCR和RT-PCR檢測(cè),獲得8株轉(zhuǎn)基因煙草植株。形態(tài)學(xué)比較表明,轉(zhuǎn)基因煙草植株與對(duì)照植株無(wú)明顯差異,說(shuō)明SmTTG1基因過(guò)量表達(dá)不一定對(duì)煙草的形態(tài)產(chǎn)生影響。
[Abstract]:Saussurea medusa Maxima, also called jellyfish snow rabbit, is a rare and endangered medicinal plant in the Qinghai-Tibet Plateau. At the same time, it is a typical alpine plant. The dense coat of the plant is the most representative morphological feature of the jellyfish Saussurea, which is an adaptive structure of the jellyfish to the extreme environment. It is of great theoretical significance to study the formation and development of the epidermal hair of Saussurea jellyfish in order to reveal the special mechanism of adaptation of alpine plants to the environment. Therefore, the plant expression vector of SmTTG1 gene was constructed by using Saussurea jellyfish as experimental material and epidermal development related gene SmTTG1 as the research object. The vector was transferred into common tobacco by Agrobacterium tumefaciens mediated by Agrobacterium tumefaciens.
