【摘要】：目的婆羅雙樹樣基因2(SALL2)是一種腫瘤抑制基因,具有抑制腫瘤細胞生長,促使腫瘤細胞凋亡的作用。檢測SALL2基因在口腔腫瘤中的表達情況,研究其對口腔腫瘤細胞增殖、遷移能力的影響,探討SALL2是否可作為口腔腫瘤標志物,在口腔腫瘤的治療過程中發(fā)揮作用。方法構建SALL2基因過表達及敲低的載體,并將載體導入到口腔腫瘤(SCC-3)細胞株中,檢測口腔腫瘤細胞株中SALL2基因的表達情況。通過MTT實驗、細胞劃痕實驗觀察細胞增殖、遷移的變化;收集15組口腔腫瘤組織與瘤旁組織,應用QRT-PCR法檢測組織中SALL2 m RNA的表達。結果1.設計并合成含有目的基因的質粒,并將質粒通過慢病毒包裝的方法導入口腔腫瘤細胞,通過此方法可構建穩(wěn)定過表達或基因敲低的口腔腫瘤細胞株;2.通過劃痕實驗結果比較各組SCC-3細胞株向劃痕中間生長移動的速度,SALL2基因的過表達具有抑制口腔腫瘤細胞遷移的作用,而敲低SALL2基因具有增強口腔腫瘤細胞遷移的作用。3.通過MTT實驗檢測各組SCC-3細胞株的生長曲線,SALL2基因的過表達具有抑制口腔腫瘤細胞增殖的作用,而敲低SALL2基因具有增強口腔腫瘤細胞增殖的作用。4.QRT-PCR法對15組標本組織中SALL2 m RNA的表達水平進行檢測,表明在口腔腫瘤組織中,SALL2的表達水平相較于正常組織呈減少趨勢。結論1.SALL2參與口腔腫瘤的發(fā)生及發(fā)展,通過一定的機制對口腔腫瘤細胞周期的調控,抑制腫瘤細胞的遷移和生長。2.SALL2可以作為口腔腫瘤的較有潛力的腫瘤分子標記物來判定腫瘤細胞的生物學行為,監(jiān)測腫瘤的轉移,評估治療效果以及預后提供新的思路。3.SALL2在口腔腫瘤組織中表達的調節(jié)機制,生物學功能以及抑制腫瘤轉移的具體機制尚未闡明,本研究為進一步探討SALL2在口腔腫瘤中的作用機制提供了一定的理論基礎。
[Abstract]:Objective Borneo double tree like gene 2pSALL2 is a tumor suppressor gene, which can inhibit the growth of tumor cells and promote apoptosis of tumor cells. To detect the expression of SALL2 gene in oral tumor, to study its effect on the proliferation and migration of oral tumor cells, and to explore whether SALL2 can be used as a tumor marker of oral cavity and play an important role in the treatment of oral tumor. Methods the overexpression and knockdown vector of SALL2 gene was constructed and introduced into oral cancer cell line SCC-3 to detect the expression of SALL2 gene in oral tumor cell line. The changes of cell proliferation and migration were observed by MTT assay and cell scratch assay, and the expression of SALL2 m RNA was detected by QRT-PCR method in 15 groups of oral tumor tissues and adjacent tissues. Result 1. The plasmid containing the target gene was designed and synthesized, and the plasmid was transferred into oral cancer cells by lentivirus packaging. By this method, stable over-expression or gene knockout of oral tumor cell line could be constructed. The results of scratch test showed that overexpression of SALL2 gene could inhibit migration of oral tumor cells, while knockout of SALL2 gene could enhance migration of oral tumor cells. The overexpression of SALL2 gene in SCC-3 cell lines was detected by MTT assay, which could inhibit the proliferation of oral cancer cells. However, knockout of SALL2 gene could enhance the proliferation of oral tumor cells. 4. The expression level of SALL2 m RNA in 15 groups was detected by QRT-PCR. The results showed that the expression of SALL2 m RNA in oral tumor tissues was decreased compared with that in normal tissues. Conclusion 1.SALL2 is involved in the occurrence and development of oral tumor and regulates the cell cycle of oral tumor through certain mechanism. Inhibiting the migration and growth of tumor cells. 2. SALL2 can be used as a potential tumor molecular marker for oral cancer to determine the biological behavior of tumor cells and to monitor tumor metastasis. 3. The regulatory mechanism of SALL2 expression in oral tumor tissues, the biological function and the specific mechanism of inhibiting tumor metastasis have not been elucidated. This study provides a theoretical basis for further study of the mechanism of SALL2 in oral tumor.
