荷花腐敗病病原菌的形態(tài)學(xué)與多基因分子系統(tǒng)學(xué)鑒定

【摘要】:荷花(Nelumbo nucifera)是我國十大名花中唯一的水生花卉,在珠三角等濕地廣泛栽培。近年來,荷花栽培集中區(qū),大范圍發(fā)生腐敗病,影響了正常的生產(chǎn)與旅游觀光產(chǎn)業(yè),為了有效控制病害發(fā)生蔓延,生產(chǎn)上急需明確荷花腐敗病的病原。本研究通過病原菌致病力測定、形態(tài)比較、分子特異檢測,以及基于翻譯延伸因子(TEF-1α)、rDNA基因內(nèi)間隔區(qū)(IGS)、線粒體小亞基(mtSSU)等基因片段的系統(tǒng)發(fā)育分析,對采自廣東、廣西、江西、福建、湖南、湖北等地的荷花腐敗病病原菌進(jìn)行鑒定及系統(tǒng)發(fā)育研究,結(jié)果如下:(1)形態(tài)學(xué)與分子鑒定均表明荷花腐敗病菌為Fusarium commune,該菌為尖孢鐮孢菌(F.oxysporum)的近緣種,首次在荷花上報道。(2)F.commune與尖孢鐮孢菌(F.oxysporum)在形態(tài)上非常相似,F.commune能產(chǎn)生長度大于25μm的細(xì)長形單瓶梗及偶爾產(chǎn)生復(fù)瓶梗的形態(tài)特征,區(qū)別于F.oxysporum。MtSSU、EF-1及IGS等基因位點(diǎn)可用于F.commune和F.oxysporum這兩個種的明確區(qū)分。(3)我國不同地區(qū)的荷花腐敗病菌存在一定的遺傳分化,荷花腐敗病菌與分離自我國的荸薺枯萎病菌(Fusarium commune)為同一個種。
[Abstract]:Nelumbo nucifera (Nelumbo nucifera) is the only aquatic flower among the ten famous flowers in China. It is widely cultivated in wetlands such as the Pearl River Delta. In recent years, there has been widespread corruption in the concentrated area of lotus cultivation, which has affected the normal production and tourism industry. In order to effectively control the spread of the disease, the pathogen of lotus corruption is urgently needed in production. In this study, pathogenicity test, morphological comparison, molecular specific detection, and phylogenetic analysis of the gene fragments from Guangdong, Guangxi, Jiangxi Province were carried out based on the intergenic spacer IGSU and mitochondrial small subunit mtSSU. In Fujian, Hunan and Hubei provinces, the pathogen of lotus rot was identified and its phylogeny was studied. The results are as follows: 1) Morphology and molecular identification indicate that the pathogen is Fusarium commune, which is a relative species of F. oxysporum. It is reported for the first time that the morphology of F. commune and F. oxysporum is very similar to that of F. oxysporum. Different from F.oxysporum.MtSSUUEF-1 and IGS, they can be used to distinguish the two species, F.commune and F.oxysporum. 3) there is a certain genetic differentiation between Lotus spoilage bacteria in different regions of China, and the same species as Fusarium commune, which is isolated from Chinese water chestnut wilt pathogen.
