鈣對(duì)染鉛大鼠海馬NMDAR和mGluR5受體介導(dǎo)相關(guān)基因mRNA及CaMKⅡ蛋白表達(dá)的影響

【摘要】:目的通過(guò)研究鈣對(duì)染鉛大鼠海馬NMDAR和m Glu R5受體介導(dǎo)相關(guān)基因mRNA及CaMKII蛋白表達(dá)的影響,為進(jìn)一步以補(bǔ)鈣預(yù)防鉛的危害提供理論依據(jù)。方法選擇體質(zhì)量(50±2)g的清潔級(jí)雄性SD大鼠60只,共分五組,每組12只,分別為對(duì)照組(基礎(chǔ)飼料,蒸餾水)、染鉛組(基礎(chǔ)飼料,0.2%醋酸鉛飲水)、試驗(yàn)Ⅰ組(基礎(chǔ)飼料添加0.5%碳酸鈣,0.2%醋酸鉛飲水)、試驗(yàn)Ⅱ組(基礎(chǔ)飼料添加1%碳酸鈣,0.2%醋酸鉛飲水)和試驗(yàn)Ⅲ組(基礎(chǔ)飼料添加2%碳酸鈣,0.2%醋酸鉛飲水)。連續(xù)飼養(yǎng)60 d后,取海馬,用熒光定量RT-PCR法測(cè)定海馬中Grm5、Camk2a、Grin1、Homer1、Prkca和ERK2的mRNA表達(dá)量,Western-blotting測(cè)定大鼠海馬中CaMKII的蛋白表達(dá)量。結(jié)果與對(duì)照組相比,染鉛組海馬中Grm5、Camk2a、Grin1、Homer1、Prkca、ERK2的mRNA表達(dá)量及CaMKII的蛋白表達(dá)量均極顯著降低(P0.01)。與染鉛組相比,試驗(yàn)組鈣濃度在一定范圍內(nèi)越高,目的基因表達(dá)量相對(duì)升高越多,試驗(yàn)Ⅱ組CaMKII的蛋白表達(dá)量極顯著上升(P0.01)。結(jié)論一定濃度的鈣可干預(yù)鉛致大鼠海馬組織的損傷。
