雙側(cè)杏仁核注射降鈣素基因相關(guān)肽對(duì)小鼠學(xué)習(xí)記憶的作用

【摘要】:本研究旨在探討外源性給予杏仁核中央核區(qū)不同濃度的降鈣素基因相關(guān)肽(calcitonin gene-related peptide,CGRP)對(duì)小鼠認(rèn)知和學(xué)習(xí)記憶的影響。將30日齡C57BL/6J小鼠隨機(jī)分為空白對(duì)照組、手術(shù)對(duì)照組、200 ng CGRP組、400 ng CGRP組和800 ng CGRP組,每組10只。外源性給予雙側(cè)杏仁核中央核區(qū)不同濃度的CGRP后,用曠場(chǎng)實(shí)驗(yàn)檢測(cè)各組小鼠認(rèn)知功能,用新物體認(rèn)知實(shí)驗(yàn)和Morris水迷宮實(shí)驗(yàn)檢測(cè)各組小鼠學(xué)習(xí)記憶能力。曠場(chǎng)實(shí)驗(yàn)結(jié)果顯示,與空白對(duì)照組相比,800 ng CGRP組小鼠的水平運(yùn)動(dòng)得分顯著增加。新物體認(rèn)知實(shí)驗(yàn)結(jié)果顯示,與空白對(duì)照組相比,400 ng CGRP組的認(rèn)知指數(shù)顯著提高。Morris水迷宮實(shí)驗(yàn)結(jié)果顯示,與空白對(duì)照組比較,400和800 ng CGRP組小鼠的潛伏期均顯著縮短,穿越平臺(tái)的次數(shù)顯著增加;與200 ng CGRP組比較,400和800 ng CGRP組小鼠尋找平臺(tái)的潛伏期均明顯縮短。以上結(jié)果提示,雙側(cè)杏仁核注射CGRP可劑量依賴性促進(jìn)小鼠非空間和空間學(xué)習(xí)記憶功能。
[Abstract]:The purpose of this study was to investigate the effects of exogenous administration of calcitonin gene-related peptidea on cognition, learning and memory of mice in the central amygdaloid nucleus region with different concentrations of calcitonin gene related peptide (calcitonin gene-related). The 30-day-old C57BL/6J mice were randomly divided into control group (n = 10), operation control group (n = 10), control group (n = 10) and control group (n = 10). After exogenous administration of different concentrations of CGRP in the central nucleus of amygdala, the cognitive function of each group was tested by open field test, and the learning and memory ability of each group was measured by new object cognition experiment and Morris water maze test. The results of open field test showed that the horizontal exercise scores of mice in the 800ng CGRP group were significantly higher than those in the blank control group. The results of new object cognition test showed that compared with the control group, the cognitive index of the 400ng CGRP group was significantly higher than that of the control group. The results showed that compared with the blank control group, the latency of mice in the 400ng CGRP group was significantly shorter than that in the control group. Compared with 200ng CGRP group, the latency of searching for platform was significantly shortened in mice of 400ng CGRP group and 800ng CGRP group. These results suggest that bilateral amygdala injection of CGRP can promote spatial and spatial learning and memory function in a dose-dependent manner.
