糖尿病大鼠P-糖蛋白編碼基因Abcb1 mRNA的表達(dá)及其對(duì)口服恩諾沙星藥動(dòng)學(xué)的影響

【摘要】:試驗(yàn)旨在研究糖尿病大鼠肝臟、腎臟、空腸和回腸中P-糖蛋白編碼基因Abcb1 mRNA的表達(dá)水平及其對(duì)口服恩諾沙星藥動(dòng)學(xué)的影響。通過鏈脲佐菌素(STZ)誘導(dǎo)建立清潔級(jí)雄性Sprague-Dawley(SD)大鼠的糖尿病模型,采用實(shí)時(shí)熒光定量法檢測(cè)并比較2組(健康組和糖尿病組)大鼠不同器官中Abcb1 mRNA的表達(dá)差異,采用高效液相色譜法(HPLC)進(jìn)一步測(cè)定糖尿病大鼠口服恩諾沙星的血藥濃度。結(jié)果表明,糖尿病大鼠肝臟和回腸中Abcb1a mRNA的表達(dá)量顯著高于健康組(P0.05),腎臟和空腸中Abcb1a mRNA的表達(dá)量雖有波動(dòng),但差異不顯著(P0.05)。健康大鼠各器官中Abcb1b mRNA的表達(dá)水平與糖尿病大鼠相比未見顯著差異(P0.05)。口服恩諾沙星的藥動(dòng)學(xué)結(jié)果表明,糖尿病組大鼠血液中恩諾沙星的峰濃度、0~12 h的藥-時(shí)曲線下面積和表觀分布容積與健康組大鼠相比,均發(fā)生顯著變化(P0.01、P0.05、P0.05),其他藥動(dòng)學(xué)參數(shù)雖有變化,但差異不顯著(P0.05)。推測(cè),糖尿病會(huì)導(dǎo)致大鼠體內(nèi)Abcb1a mRNA的表達(dá)量發(fā)生變化,進(jìn)一步導(dǎo)致口服藥物在大鼠體內(nèi)藥動(dòng)學(xué)過程發(fā)生變化。
[Abstract]:The aim of this study was to investigate the expression of Abcb1 mRNA in liver, kidney, jejunum and ileum of diabetic rats and its effect on oral enrofloxacin pharmacokinetics. The diabetic model of clean male Sprague-Dawley SD rats was established by streptozotocin (STZ). The expression of Abcb1 mRNA in different organs of two groups (healthy group and diabetic group) was detected and compared by real-time fluorescence quantitative method. The serum concentration of enrofloxacin in diabetic rats was further determined by HPLC. The results showed that the expression of Abcb1a mRNA in liver and ileum of diabetic rats was significantly higher than that in healthy control group (P 0.05). Although the expression of Abcb1a mRNA in kidney and jejunum fluctuated, the difference was not significant. There was no significant difference in the expression of Abcb1b mRNA between healthy rats and diabetic rats (P 0.05). The pharmacokinetic results of oral enrofloxacin showed that the peak concentration of enrofloxacin in blood of diabetic rats was less than that of healthy rats. There were significant changes in P0.01P0.05P0.05 and other pharmacokinetic parameters, but the difference was not significant. It is speculated that diabetes may cause changes in the expression of Abcb1a mRNA in rats and further changes in pharmacokinetics of oral drugs in rats.
