貴州漢族人群載脂蛋白B基因EcoRI位點(diǎn)多態(tài)性與原發(fā)性高血壓相關(guān)性的研究

【摘要】:目的:研究貴州地區(qū)漢族人群載脂蛋白B(apoB)基因EcoRI位點(diǎn)多態(tài)性與原發(fā)性高血壓(EH)的關(guān)系,并探討該位點(diǎn)的多態(tài)性與血脂及其他心血管危險(xiǎn)因素的關(guān)系。方法:采用聚合酶鏈反應(yīng)-限制性片段長(zhǎng)度多態(tài)性的方法分別檢測(cè)81例高血壓患者及71例血壓正常者的apoB基因EcoRI位點(diǎn)的多態(tài)性,比較該位點(diǎn)不同基因型的頻率、等位基因的頻率,并對(duì)血脂、體重指數(shù)(BMI)、肌酐、尿素氮、糖化血紅蛋白(HbAlc)等指標(biāo)進(jìn)行測(cè)定。結(jié)果:(1)在正常對(duì)照組中,apoB基因EcoRI位點(diǎn)的E+E+和E+E-基因型頻率分別為0.887和0.113;在高血壓組中,EcoRI位點(diǎn)的E+E+、E+E-基因型頻率分別為0.864和0.136;兩組均未檢測(cè)到E-E-基因型;經(jīng)比較,未發(fā)現(xiàn)E+E+和E+E-基因型頻率在兩組間的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(2)正常組中E+和E-等位基因的頻率分別為0.944和0.056;高血壓組中E+和E-等位基因的頻率分別為0.932和0.068;經(jīng)比較,兩組在該位點(diǎn)的等位基因頻率的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。(3)在EH組中,攜帶E+E-基因型個(gè)體的HbAlc和低密度脂蛋白膽固醇(LDL-C)水平顯著高于攜帶E+E+基因型個(gè)體,而B(niǎo)MI、肌酐、尿素氮、甘油三酯(TG)、總膽固醇(TC)、高密度脂蛋白膽固醇(HDL-C)、LDL-C/HDL-C比值、TC/HDL-C比值以及脂蛋白a[LP(a)]等在兩組中EcoRI位點(diǎn)基因型頻率的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);在正常對(duì)照組中,上述指標(biāo)與EcoRI位點(diǎn)的多態(tài)性無(wú)明顯的相關(guān)性(P0.05)。結(jié)論:本研究發(fā)現(xiàn)apoB基因EcoRI位點(diǎn)多態(tài)性與貴州漢族人群原發(fā)性高血壓的發(fā)生無(wú)關(guān),但該位點(diǎn)的多態(tài)性與高血壓患者HbAlc和LDL-C水平有關(guān)。
[Abstract]:Objective: to study the relationship between EcoRI polymorphism of apolipoprotein (apoB) gene and essential hypertension (EH) in Guizhou Han population, and to explore the relationship between polymorphism of ApoB gene and blood lipids and other cardiovascular risk factors. Methods: polymerase chain reaction-restriction fragment length polymorphism (PCR) was used to detect the polymorphism of apoB gene EcoRI locus in 81 patients with hypertension and 71 patients with normal blood pressure, and the frequencies of different genotypes of this locus were compared. The frequency of alleles and serum lipids, body mass index (BMI), creatinine, urea nitrogen, HbAlc were measured. Results the frequencies of E E and E E-genotype at EcoRI site of apoB gene were 0.887 and 0.113 in normal control group, 0.864 and 0.136 in hypertensive group, respectively. The frequencies of E and E- alleles in normal controls were 0.944 and 0.056 respectively, and the frequencies of E and E- alleles in hypertension group were 0.932 and 0.068, respectively. In EH group, the levels of HbAlc and LDL-C in E- genotype individuals were significantly higher than those in E E genotype individuals, while BMI, creatinine, urea nitrogen were significantly higher in EH group. There was no significant difference in the genotype frequency of EcoRI locus between the two groups, such as triglyceride, total cholesterol, HDL-C / HDL-C ratio and lipoprotein a, in the two groups; in the normal control group, there was no significant difference in the genotypic frequencies of EcoRI locus between the two groups (P 0.05), but in the normal control group, there was no significant difference in the genotype frequency of EcoRI locus between the two groups. There was no significant correlation between the above indexes and the polymorphism of EcoRI locus (P 0.05). Conclusion: this study found that the polymorphism of EcoRI locus of apoB gene was not related to the occurrence of essential hypertension in Guizhou Han population, but the polymorphism of this locus was related to the levels of HbAlc and LDL-C in patients with hypertension.
