新疆南疆驢源H3N8亞型EIVHA基因序列分析

【摘要】:為明確新疆南疆驢是否感染馬流感病毒(Equine influenza virus,EIV)及驢源EIV HA基因特征,根據(jù)GenBank登錄的EIV基因序列,設(shè)計(jì)合成1對(duì)引物,采集新疆南疆地區(qū)2個(gè)驢屠宰點(diǎn)33份驢肺組織樣品,采用RT-PCR擴(kuò)增驢源EIV HA基因片段,并對(duì)擴(kuò)增產(chǎn)物進(jìn)行測(cè)序與序列分析。結(jié)果表明,中國(guó)新疆南疆驢源EIV A/donkey/Xinjiang/1/2015(H3N8)株HA基因長(zhǎng)1 704bp,編碼567個(gè)氨基酸。同源性分析結(jié)果顯示,與國(guó)內(nèi)外H3N8亞型EIV參考毒株核苷酸和氨基酸同源性分別為88.9%~99.9%和88.3%~99.8%,與國(guó)內(nèi)毒株和哈薩克斯坦毒株在一個(gè)進(jìn)化分支內(nèi)。與GenBank數(shù)據(jù)庫(kù)中唯一1個(gè)驢源毒株A/donkey/Xinjiang/5/2007(H3N8)的比對(duì)結(jié)果顯示,2個(gè)毒株裂解位點(diǎn)、糖基化位點(diǎn)、抗原位點(diǎn)和受體結(jié)合位點(diǎn)氨基酸完全一致,只是錯(cuò)位2個(gè)氨基酸。確定新疆南疆存在驢感染EIV,可能為國(guó)內(nèi)疫情的延續(xù)或周邊國(guó)家傳入。
[Abstract]:In order to find out whether the donkeys are infected with equine influenza virus and the EIV HA gene of donkeys in southern Xinjiang, a pair of primers were designed and synthesized according to the EIV gene sequence registered by GenBank, and 33 donkey lung tissue samples were collected from two donkey slaughtering sites in southern Xinjiang. Donkey EIV HA gene fragment was amplified by RT-PCR and sequenced. The results showed that the HA gene of the donkeys' EIV A / R / Xinjiang / 1 / 2015 H3N8 strain was 1704 BP and encoded 567 amino acids. The results of homology analysis showed that the nucleotide and amino acid homology of the reference strains of H3N8 subtype EIV were 88.999% and 88.3% respectively, which were in an evolutionary branch with domestic strains and Kazakstan strains. The results of comparison with the only donkey virus strain A / R / Xinjiangr / 5 / 2007 H3N8 in GenBank database showed that the amino acids of the two strains were identical in cleavage site, glycosylation site, antigen site and receptor binding site, but only 2 amino acids were translocated. It is confirmed that the existence of donkey infection in southern Xinjiang may be a continuation of the domestic epidemic situation or the introduction of neighboring countries.
