【摘要】：目的:探討LKB1基因表達(dá)水平與NSCLC患者預(yù)后因素的相關(guān)性。方法:選取2015年2月至2016年8月弋磯山醫(yī)院胸外科手術(shù)切除NSCLC癌組織石蠟包埋標(biāo)本、新鮮組織標(biāo)本23例為肺癌組,所有標(biāo)本均經(jīng)病理學(xué)檢測(cè),確診為肺鱗癌、腺癌。同時(shí)選取同一患者癌旁正常肺部組織23例作為空白對(duì)照組。使用RT-PCR和Western-blot法檢測(cè)癌組織及癌旁組織中LKB1基因mRNA和蛋白的表達(dá)量,并分析其與年齡、分化程度、臨床分期、腫瘤大小、無(wú)進(jìn)展生存期的關(guān)系。結(jié)果:在肺癌組,LKB1基因表達(dá)低于空白對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P0.05),LKB1表達(dá)水平與患者年齡、臨床分期、腫瘤大小無(wú)相關(guān)性(P0.05),與分化程度呈正相關(guān)(P0.05)。LKB1 mRNA高表達(dá)組肺癌患者中位無(wú)進(jìn)展生存期為13.033月,低表達(dá)組中位生存期為5.267月,高表達(dá)組無(wú)進(jìn)展生存期為(13.247±0.505)個(gè)月,低表達(dá)組無(wú)進(jìn)展生存期為(6.426±1.251)個(gè)月,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。LKB1蛋白高表達(dá)組肺癌患者中位生存期為12.833個(gè)月,低表達(dá)組中位生存期為8.467個(gè)月,LKB1蛋白高表達(dá)組無(wú)進(jìn)展生存期為(13.487±1.079)個(gè)月,低表達(dá)組無(wú)進(jìn)展生存期為(8.239±0.979)個(gè)月,差異有統(tǒng)計(jì)學(xué)意義(P0.05).結(jié)論:LKB1基因在非小細(xì)胞肺癌中的表達(dá)明顯低于相應(yīng)的癌旁組織,LKB1表達(dá)量與非小細(xì)胞肺癌分化程度相關(guān),且LKB1低表達(dá)可能提示預(yù)后不良。
[Abstract]:Objective: to investigate the correlation between LKB1 gene expression and prognostic factors in NSCLC patients. Methods: paraffin embedded specimens of NSCLC cancer tissue were removed by thoracic surgery from February 2015 to August 2016. 23 cases of lung cancer were collected from fresh tissues. All the specimens were diagnosed as squamous cell carcinoma and adenocarcinoma by pathology. At the same time, 23 cases of normal lung tissue adjacent to cancer of the same patient were selected as blank control group. RT-PCR and Western-blot methods were used to detect the expression of LKB1 gene mRNA and protein in cancer tissues and adjacent tissues, and to analyze the relationship between LKB1 gene mRNA and protein expression and age, differentiation, clinical stage, tumor size and progression free survival. Results: the expression of LKB1 gene in lung cancer group was lower than that in blank control group. There was no correlation between tumor size and differentiation degree. The median progressive survival time of lung cancer patients with high expression of P0.05U. LKB1 mRNA was 13.03 3 months, the median survival time of low expression group was 5.267 months, and that of high expression group was 13.247 鹵0.505 months. The progression free survival time of low expression group was 6.426 鹵1.251 months, the median survival time of lung cancer patients with high expression of P0.05U. LKB1 protein was 12.833 months, the median survival time of low expression group was 8.467 months and that of high expression group of LKB1 protein was 13.487 鹵1.079 months. The progression-free survival time of low expression group was 8.239 鹵0.979 months, and the difference was statistically significant (P 0.05). Conclusion the expression of the LKB1 gene in non-small cell lung cancer is significantly lower than that in the adjacent tissues of non-small cell lung cancer. The low expression of LKB1 may indicate that the prognosis of NSCLC is poor.
