下調(diào)STYK1基因表達(dá)可抑制結(jié)直腸癌細(xì)胞生長與侵襲

【摘要】:目的探討STYK1基因下調(diào)對結(jié)直腸癌細(xì)胞HCT-15增殖、凋亡和遷移以及MEK/ERK和PI3K/AKT信號通路的影響。方法采用基因干擾技術(shù)抑制結(jié)直腸癌細(xì)胞HCT-15中STYK1的表達(dá),Western blot檢測抑制效果,CCK-8檢測STYK1表達(dá)下調(diào)對HC-T15細(xì)胞增殖的影響,流式細(xì)胞儀檢測STYK1表達(dá)下調(diào)對HC-T15細(xì)胞凋亡的影響,Transwell小室實(shí)驗(yàn)檢測STYK1表達(dá)下調(diào)對HCT-15細(xì)胞侵襲的影響,通過Western blot檢測STYK1表達(dá)下調(diào)對HCT-15細(xì)胞MEK/ERK和PI3K/AKT信號通路活性的影響。結(jié)果與干擾對照組相比,STYK1表達(dá)下調(diào)明顯抑制HCT-15細(xì)胞增殖活性(P0.01)和侵襲能力(P0.01),并誘導(dǎo)細(xì)胞凋亡(P0.01);同時(shí),STYK1表達(dá)下調(diào)明顯抑制HCT-15細(xì)胞ERK1/2和AKT蛋白的磷酸水平。結(jié)論 STYK1基因干擾可以抑制結(jié)直腸癌細(xì)胞的增殖和侵襲,并誘導(dǎo)凋亡,其機(jī)制可能與抑制MEK/ERK和PI3K/AKT信號通路活性有關(guān)。
[Abstract]:Objective to investigate the effects of down-regulation of STYK1 gene on HCT-15 proliferation, apoptosis and migration, MEK/ERK and PI3K/AKT signaling pathway in colorectal cancer cells. Methods Gene interference technique was used to inhibit the expression of STYK1 in HCT-15 of colorectal cancer cells. Western blot was used to detect the inhibitory effect of CCK-8 on the proliferation of HC-T15 cells. Effect of down-regulation of STYK1 expression on apoptosis of HC-T15 cells the effect of down-regulation of STYK1 expression on the invasion of HCT-15 cells was detected by flow cytometry, and the effect of down-regulation of STYK1 expression on MEK/ERK and PI3K/AKT signaling pathway activity in HCT-15 cells was detected by Western blot. Results compared with the control group, the down-regulation of STYK1 expression significantly inhibited the proliferation activity of HCT-15 cells (P0.01) and the invasion ability of P0.01G, and induced apoptosis (P0.01G), while the down-regulation of STYK1 expression significantly inhibited the phosphoric acid levels of ERK1/2 and AKT proteins in HCT-15 cells. Conclusion STYK1 gene interference can inhibit the proliferation and invasion of colorectal cancer cells and induce apoptosis. The mechanism may be related to the inhibition of MEK/ERK and PI3K/AKT signaling pathway activity.
