大腸癌圍手術(shù)期患者陰虛證差異表達(dá)基因及信號(hào)通路分析

【摘要】:方法:針對(duì)大腸癌圍手術(shù)期患者的主要中醫(yī)證候陰虛證,采集符合條件病例的大腸癌組織及癌周正常組織,采用羅氏公司含12×135K個(gè)基因的人全基因組芯片檢測(cè)大腸癌組織差異表達(dá)基因,篩選并分析陰虛證癌組織組和癌周正常組織組的差異表達(dá)基因,利用Pathway對(duì)相關(guān)基因進(jìn)行生物信息學(xué)分析。結(jié)果:得出大腸癌圍手術(shù)期患者陰虛證癌組織組和癌周正常組織組表達(dá)的差異基因共4869個(gè),包括上調(diào)基因1574個(gè),下調(diào)基因3295個(gè)。經(jīng)檢索數(shù)據(jù)庫(kù)Gene Bank,獲取基因名稱等相關(guān)信息。進(jìn)行Pathway分析,篩選出陰虛證癌組織組和癌周正常組織組之間具有顯著差異表達(dá)基因主要涉及的信號(hào)轉(zhuǎn)導(dǎo)通路。結(jié)論:大腸癌圍手術(shù)期陰虛證癌組織組與癌周正常組織組比較,存在組織差異基因表達(dá)特征圖譜,主要涉及主要涉及炎癥反應(yīng),細(xì)胞內(nèi)吞功能,礦物質(zhì)吸收,酮體合成與分解功能,免疫功能等相關(guān)基因。其主要信號(hào)轉(zhuǎn)導(dǎo)通路包括MAPK信號(hào)通路及p53信號(hào)通路等。
[Abstract]:Methods: according to the main syndromes of yin deficiency in patients with colorectal cancer during perioperative period, the tissues of colorectal cancer and normal tissues around cancer were collected. The differentially expressed genes in colorectal cancer tissues were detected by human genome microarray with 12 脳 135K genes in Roche Company. The differentially expressed genes were screened and analyzed in the cancer tissues of Yin-deficiency syndrome and the normal tissues around cancer. The related genes were analyzed by bioinformatics using Pathway. Results: there were 4869 differentially expressed genes including 1574 up-regulated genes and 3295 down-regulated genes. The database Gene Bank was searched for gene name and other related information. By Pathway analysis, the signal transduction pathways involved in the significant difference of expression genes between cancer tissues of yin deficiency syndrome and normal tissues around carcinoma were screened. Conclusion: there is a characteristic map of differentially expressed genes in the tissues of cancer with deficiency of yin and normal tissues around carcinoma of large intestine in perioperative period, which mainly involves inflammation, endocytosis and mineral absorption. Ketone synthesis and decomposition function, immune function and other related genes. Its main signal transduction pathways include MAPK signaling pathway and p53 signaling pathway.
